To a solution of methyl 3-cyclohexyl-2-(1,2-dihydroxyethyl)-1-[2-(dimethylamino)-2-oxoethyl]-1H-indole-6-carboxylate (from Step 3) in THF/water (0.05 M, 1:1, v:v) cooled to 0° C. was added sodium periodate (1.2 eq.). The resulting mixture was allowed to warm to RT and left stirring for a further 3 h. The mixture was diluted with EtOAc and 5% aq. citric acid solution. The phases were separated and the aqueous phase was extracted with EtOAc. The combined organic phases were extracted with aqueous citric acid (2% solution), sat. aq. NaHCO3 solution and brine. The organic phase was then dried over Na2SO4 and evaporated in vacuo. The residual material was then crystallised from EtOAc with PE. The product was obtained as an off-white solid (83%).
