Non-Human Toxicity Excerpts
/ALTERNATIVE and IN VITRO TESTS/ The preparation of R-(-)-3-quinuclidinol and its acetate ester is reported. These compounds, together with their racemates, were tested as inhibitors and substrates of the enzyme acetylcholinesterase. The results suggest that the enzyme active site is stereospecific toward R-(-)-3-quinuclidinol hydrochloride when acting as an inhibitor. Both (+/-)- and R-(-)-3-acetoxyquinuclidine hydrochloride are substrates of the enzyme, but the S-enantiomer probably has some affinity for the enzyme also.
/ALTERNATIVE and IN VITRO TESTS/ Choline uptake into cholinergic neurons for acetylcholine (ACh) synthesis is by a specific, high-affinity, sodium- and temperature-dependent transport mechanism (HAChU). To assess the role of choline availability in regulation of ACh synthesis, the structure-activity relationships of several ... quinuclidinyl analogs were evaluated in a dose response manner. ... The most potent quinuclidinyl derivative was the N-methyl-3-quinuclidinone (I50 = 5.6 X 10-7 M). This compound had approximately 100-fold greater inhibitory activity than the corresponding racemic alcohol, suggesting that the 3-hydroxyl functional group is not absolutely essential for activity. Increasing the size of the N-functional group from a methyl to an allyl in the alcohol led to a 10-fold increase in activity. However, removal of the quaternizing N-methyl group yielding the tertiary amine, 3-quinuclidinol hydrochloride, greatly reduced its capacity to inhibit HAChU. Of the 2-benzylidene-3-quinuclidinone derivatives studied, only the m-chloro derivative significantly reduced HAChU. /3-Quinuclidinol hydrochloride/
Environmental Abiotic Degradation
The rate constant for the vapor-phase reaction of 3-quinuclidinol with photochemically-produced hydroxyl radicals has been estimated as 5.5X10-11 cu cm/molecule-sec at 25 °C(SRC) using a structure estimation method(1). This corresponds to an atmospheric half-life of about 2.3 hours at an atmospheric concentration of 5X10+5 hydroxyl radicals per cu cm(1). 3-Quinuclidinol is not expected to undergo hydrolysis in the environment due to the lack of functional groups that hydrolyze under environmental conditions(3). 3-Quinuclidinol does not contain chromophores that absorb at wavelengths >290 nm(3) and therefore is not susceptible to direct photolysis by sunlight(SRC).
Environmental Fate/Exposure Summary
3-Quinuclidinol's production and use as a synthetic precursor to chemical warfare agents and drugs may result in its release to the environment through various waste streams. If released to air, an estimated vapor pressure of 3.3X10-4 mm Hg at 25 °C indicates 3-quinuclidinol will exist solely as a vapor in the atmosphere. Vapor-phase 3-quinuclidinol will be degraded in the atmosphere by reaction with photochemically-produced hydroxyl radicals; the half-life for this reaction in air is estimated to be 2.3 hours. 3-Quinuclidinol does not contain chromophores that absorb at wavelengths >290 nm and therefore is not expected to be susceptible to direct photolysis by sunlight. If released to soil, 3-quinuclidinol is expected to have very high mobility based upon an estimated Koc of 8. Volatilization from moist soil surfaces is not expected to be an important fate process based upon an estimated Henry's Law constant of 8.1X10-10 atm-cu m/mole. 3-Quinuclidinol is not expected to volatilize from dry soil surfaces based upon its vapor pressure. If released into water, 3-quinuclidinol is not expected to adsorb to suspended solids and sediment based upon the Koc. Volatilization from water surfaces is not expected to be an important fate process based upon Henry's Law constant of 3-quinuclidinol. An estimated BCF of 3.2 suggests the potential for bioconcentration in aquatic organisms is low. Hydrolysis is not expected to be an important environmental fate process since 3-quinuclidinol lacks functional groups that hydrolyze under environmental conditions. Occupational exposure to 3-quinuclidinol may occur through inhalation of dust and dermal contact at workplaces where it is produced or used. (SRC)
