| Gefahrenübersicht | Malathion is an insecticide that does not occur naturally. Pure malathion is a colorless liquid, and technical-grade malathion, which contains >90% malathion and impurities in a solvent, is a brownish-yellow liquid that smells like garlic. Malathion is used to kill insects on farm crops and in gardens, to treat lice on humans, and to treat fleas on pets. Malathion is also used to kill mosquitos and Mediterranean fruit flies on (medflies) in large outdoor areas.Malathion is one of the less potent organophosphate insecticides. It has an oral LD50 of 500-1000 mg/kg compared to an LD50 of 1-5 mg/kg for parathion. [LaDou, p. 584] Allergic contact dermatitis has been reported in farmers. [Kanerva, p. 1808] Inhalation is not a significant route of exposure to malathion at ordinary temperatures because of its low volatility, but toxic effects can occur after inhalation of malathion sprays or dusts. [ATSDR Medical Management] The average of two baseline respective cholinesterase activity determinations three days apart, with no exposures to enzyme inhibiting pesticides for at least 30 days, is recommended for each worker prior to exposure to cholinesterase inhibitors because of large inter-individual differences in published baseline values. To be established at least once a year. Removal from workplace exposures is recommended until the cholinesterase activity returns to within 20% of baseline. [TLVs and BEIs] |

| Körperbelastung | According to a national survey performed in the US from 1976-1980, malathion was detected in 1.6% of the urine analyses for persons 12-74 years of age(1). Trace amounts of metabolites in urine showed 4.1% of the persons tested were exposed to malathion(2). Of 267 samples of human urine, 0.4% were positive for malathion, at concentrations <0.1 ppm(3). The malathion metabolite, malathion dicarboxylic acid, was detected in 43% of 88, 29% of 89 and 36% of 89 urine samples after 3, 5 and 7 days, respectively, in samples collected for the Minnesota Children's Pesticide Exposure Survey (MNCPES)(4). Malathion was detected in 38, 27 and 32 out of 89 urine samples of children at levels of 0.3-5.6, 0.1-17.0 and 0.4-26.1 ug/g creatinine, respectively, during the MNCPES(5). Urinary metabolites of malathion were detected in 17 of 60 urine samples from 60 children 1-6 years old in eastern North Carolina farm-worker households at a median of 0.33 ug/g creatininesamples were collected 2004(6).Malathion was detected in one of 57 personal air samples at a concentration of 16.1 ng/cu m, samples were collected over a 48 hour period from the mother during the 32nd week of pregnancy, mothers reside in New York City, NY(1). Malathion was detected, not quantified in the respired air of a resident in 1 of 9 residents from 9 households sampled in Jacksonville, FL(6). |
| Treatment | If the compound has been ingested, rapid gastric lavage should be performed using 5% sodium bicarbonate. For skin contact, the skin should be washed with soap and water. If the compound has entered the eyes, they should be washed with large quantities of isotonic saline or water. In serious cases, atropine and/or pralidoxime should be administered. Anti-cholinergic drugs work to counteract the effects of excess acetylcholine and reactivate AChE. Atropine can be used as an antidote in conjunction with pralidoxime or other pyridinium oximes (such as trimedoxime or obidoxime), though the use of '-oximes' has been found to be of no benefit, or possibly harmful, in at least two meta-analyses. Atropine is a muscarinic antagonist, and thus blocks the action of acetylcholine peripherally. |
| Wechselwirkungen | An important characteristic of malathion acute toxicity is its potentiation by other organophosphates. When malathion and EPN were administered to rats separately, LD50s were 1400 and 65 mg/kg, respectively. However, when malathion and EPN were administered simultaneously at a ratio of about 25:1, oral LD50s were reduced to 167 and 7 mg/kg, respectively. The onset of symptoms for individual compounds was slow, and death usually occurred several hours after administration. However, when given together at or near the LD50, symptoms developed much more rapidly, and death usually occurred within 1 hour. In dogs, oral doses of 2000 or 4000 mg/kg malathion alone were not fatalhowever, when EPN was given simultaneously (2 or 5 mg/kg) with malathion, doses of 50, 100, and 200 mg/kg were lethal. Potentiation of malathion toxicity is due to the inhibition of carboxyesterase by EPN (and other organophosphates), an enzyme important in detoxifying of malathion (as well as the more toxic metabolic product, malaoxon). This study was performed to investigate the antidepressant-like effect of minocycline in mice exposed to organophosphate pesticide malathion and possible involvement of nitric oxide/cGMP pathway in this paradigm. Mice were administered specific doses of malathion once daily for 7 consecutive days. After induction of depression, different doses of minocycline were daily injected alone or combined with non-specific NOS inhibitor, L-NAME, specific inducible NOS inhibitor, AG, NO precursor, L-arginine, and PDE5I, sildenafil. After locomotion assessment in open-field test, immobility times were recorded in the FST and TST. Moreover, hippocampal nitrite concentrations and acetylcholinesterase activity were measured. The results showed that repeated exposure to malathion induces depressive-like behavior at a dose of 250 mg/kg. Minocycline (160 mg/kg) significantly reduced immobility times in FST and TST (P < 0.001). Combination of sub-effective doses of minocycline (80 mg/kg) with either L-NAME (3 mg/kg) or AG (25 mg/kg) significantly exerted a robust antidepressant-like effect in FST and TST (P < 0.001). Furthermore, minocycline at the same dose which has antidepressant-like effect, significantly reduced hippocampal nitrite concentration. The investigation indicates the essential role for NO/cGMP pathway in malathion-induced depressive-like behavior and antidepressant-like effect of minocycline. Moreover, the interaction between nitrergic and cholinergic systems are suggested to be involved in malathion-induced depression. It was established in experiments on noninbred albino rats that acute intoxication with malathion (0.75 LD50) reduced the function of Th1 cells more significantly than the function of Th2 lymphocyte, decreases the activity of B cells and NK cells, blood levels of TNFa, IL-1b and IL-6, IFN-g, IL-2, and IL-4, while not significantly affecting the concentration of IL-10 and IL-13. Atropine (10 mg/kg) under conditions of acute malathion intoxication improved the function of T cells and B lymphocytes, NK cells, as well as the synthesis of immunoregulatory cytokines IFN-g, IL-2, and IL-4. At the same time, atropine in malathion intoxicated rats had no effect on suppression of the synthesis of proinflammatory cytokines TNF, IL-1g and IL-6 as well as the content of anti-inflammatory cytokines IL-10 and IL-13.In this study the effect of crocin, a carotenoid isolated from saffron, on malathion (an organophosphate insecticide) induced depressive- like behavior in subacute exposure was investigated. Moreover the molecular mechanism of malathion induced depressive- like behavior and its decreasing effect on the level of brain derived neurotrophic factor (BDNF) in rat hippocampus and cerebral cortex were evaluated. Male Wistar rats were exposed to malathion (50 mg/kg/day, IP) alone or in combination with crocin (10, 20 and 40 mg/kg/day, IP), imipramine (20 mg/kg/day, IP) and vitamin E (200 mg/kg, three times a week, IP) respectively for 14 days. The forced swimming test (FST) was performed on days 1, 7 and 14. The level of malondealdehyde (MDA) and reduced glutathione (GSH) were measured in cerebral cortex and hippocampus of rats. The protein level of BDNF was evaluated using Western blot analysis. Malathion (50 mg/kg, IP) increased immobility time in the FST, without affecting total locomotor activity in open-field test. Malathion increased the malondealdehyde (MDA) and decreased the glutathione (GSH), whereas these effects were reversed by crocin and vitamin E. Malathion decreased plasma acetylcholinesterase activity, however this effect was not reversed by crocin or vitamin E. Malathion reduced the protein level of BDNF in rat hippocampus. Imipramine and crocin prevented the decreasing effect of malathion on BDNF. These results showed that crocin attenuates some neurochemical and behavioral effects induced by malathion. This neuroprotective effect of crocin may be in part due to its effect on BDNF.For more Interactions (Complete) data for MALATHION (13 total), please visit the HSDB record page. |
| Zielorgane | Neurological (Nervous System), Respiratory (From the Nose to the Lungs)Eyes, skin, respiratory system, liver, blood cholinesterase, central nervous system, cardiovascular system, gastrointestinal tract |
| Gesundheitliche Auswirkungen | Acute exposure to cholinesterase inhibitors can cause a cholinergic crisis characterized by severe nausea/vomiting, salivation, sweating, bradycardia, hypotension, collapse, and convulsions. Increasing muscle weakness is a possibility and may result in death if respiratory muscles are involved. Accumulation of ACh at motor nerves causes overstimulation of nicotinic expression at the neuromuscular junction. When this occurs symptoms such as muscle weakness, fatigue, muscle cramps, fasciculation, and paralysis can be seen. When there is an accumulation of ACh at autonomic ganglia this causes overstimulation of nicotinic expression in the sympathetic system. Symptoms associated with this are hypertension, and hypoglycemia. Overstimulation of nicotinic acetylcholine receptors in the central nervous system, due to accumulation of ACh, results in anxiety, headache, convulsions, ataxia, depression of respiration and circulation, tremor, general weakness, and potentially coma. When there is expression of muscarinic overstimulation due to excess acetylcholine at muscarinic acetylcholine receptors symptoms of visual disturbances, tightness in chest, wheezing due to bronchoconstriction, increased bronchial secretions, increased salivation, lacrimation, sweating, peristalsis, and urination can occur. Certain reproductive effects in fertility, growth, and development for males and females have been linked specifically to organophosphate pesticide exposure. Most of the research on reproductive effects has been conducted on farmers working with pesticides and insecticdes in rural areas. In females menstrual cycle disturbances, longer pregnancies, spontaneous abortions, stillbirths, and some developmental effects in offspring have been linked to organophosphate pesticide exposure. Prenatal exposure has been linked to impaired fetal growth and development. Neurotoxic effects have also been linked to poisoning with OP pesticides causing four neurotoxic effects in humans: cholinergic syndrome, intermediate syndrome, organophosphate-induced delayed polyneuropathy (OPIDP), and chronic organophosphate-induced neuropsychiatric disorder (COPIND). These syndromes result after acute and chronic exposure to OP pesticides. |

| Umweltverhalten | TERRESTRIAL FATE: Based on a classification scheme(1), Koc values of 927-17,620(2), indicate that malathion is expected to have low to no mobility in soil(SRC). Volatilization of malathion from moist soil surfaces is not expected to be an important fate process(SRC) given a Henry's Law constant of 4.89X10-9 atm-cu m/mole(3). Malathion is not expected to volatilize from dry soil surfaces(SRC) based upon a vapor pressure of 3.97X10-5 mm Hg at 30 °C(4). The degradation half-life of malathion in five different soils was 1.33-4.14 days(5). Half-lives for malathion for radish and carrot soil for the seasons of winter, summer, and post-monsoon were determined to be 6.4, 2.1, and 5.3 days, and 6.6, 2.6, and 5.6 days, respectively(6). Malathion levels found in Southern California after a single spraying event at soil depths of 1 and 0.1 cm were 1.4 ug/g and 14.1 ug/g, respectively(7). Malathion is rapidly degraded in soils with reported degradation in 10 days in various non sterile loam soils of 81-94%, degradation in the same loam soils that were sterile were 5-19%(8).TERRESTRIAL FATE: The degradation half-life of malathion was reported for five different soils in China(1).[Table#1931]AQUATIC FATE: Based on a classification scheme(1), Koc values of 927-17,620(2), indicate that malathion is expected to adsorb to suspended solids and sediment(SRC). Volatilization from water surfaces is not expected(3) based upon a Henry's Law constant of 4.89X10-9 atm-cu m/mole(4). Malathion did not bioconcentrate in the freshwater fish topmouth gudgeon (Pseudorasbora parva)(5) or in pinfish (Lagodon sp)(6). Hydrolysis half-life for malathion in seawater/sediment systems at pH 7.3-7.7 is 2.0 days and in freshwater at pH 7.4 and 20 °C is 11 days(7). Products of hydrolysis include malaoxon, malathion alpha and beta monoacid, O,O-dimethylphosphorodithioic acid, diethyl fumarate, diethyl thiomalate, O,O-dimethylphosphorothionic acid(8). Hydrolysis is the main route of malathion degradation in water with pH >7.0at pH <7.0, the rate of hydrolysis is slow relative to the rate of biodegradation(9). Photodegradation may also compete with hydrolysis based on a study finding the first order photolysis rate of malathion in river water as 0.0013/min, this calculates to a half-life of 533 minutes(10). Malathion sorbed by algae was photodegraded more than 25 times faster than malathion in distilled water(11). The half-life for malathion in Limon River water was reported as 10.87, 10.41, 9.73 and 10.50 days under respective experimental conditions of filtered-dark, filtered-sunlight, non-filtered open to the air and non-filtered closed(12). The degradation half-life of malathion in different types of waters was 1.03-3.93 days(13). Malathion proposed degradation pathway in river water proceeds via hydrolysis to O,O-dimethyldithiophosphoric acid followed by oxidation to O,O-dimethylthiophosporic acid(14).AQUATIC FATE: The degradation half-life (T1/2) and first-order degradation rate constant (k/day) of malathion were reported for five different waters in China(1).[Table#1932]ATMOSPHERIC FATE: According to a model of gas/particle partitioning of semivolatile organic compounds in the atmosphere(1), malathion, which has a vapor pressure of 3.97X10-5 mm Hg at 30 °C(2), will exist in both the vapor and particulate phases in the ambient atmosphere. Vapor-phase malathion is degraded in the atmosphere by reaction with photochemically-produced hydroxyl radicals(SRC)the half-life for this reaction in air is estimated to be 5 hours(SRC), calculated from its rate constant of 7.7X10-11 cu cm/molecule-sec at 25 °C(SRC) that was derived using a structure estimation method(3). Particulate-phase malathion may be removed from the air by wet or dry deposition(SRC). Malathion absorbs light at wavelengths >290 nm(4) and, therefore, may be susceptible to direct photolysis by sunlight(SRC). |

| Expositionspfade | The substance can be absorbed into the body by inhalation, through the skin and by ingestion.inhalation, skin absorption, ingestion, skin and/or eye contactMalathion in an acetone vehicle has been reported to be absorbed through normal human skin only to the extent of 8% of the applied dose. Absorption may be increased when malathion is applied to damaged skin. Malathion is rapidly and effectively absorbed by practically all routes including the gastrointestinal tract, skin, mucous membranes, and lungs. However, it is readily excreted in the urine, and does not accumulate in organs or tissues. |
| Milk Concentrations | Malathion was detected in 2 of 96 samples collected from 4 brands of Mexican commercial pasteurized milk, sampled over a 12 month period(1). Malathion was not detected (detection limit 0.01 mg/kg) in 20 milk samples from the market in Beijing, China(2). Malathion was detected in some of the 312 bovine milk samples from Punjab, India(3). |

Beschreibung
Malathion, a clear amber liquid, is sparingly soluble in water but soluble in a majority of organic solvents. The U.S. Environmental Protection Agency (U.S. EPA) grouped malathion under GUP, meaning general use pesticide. It is used as an insecticide as well as an acaricide for the control of pests. Malathion is used for the control of sucking insects and chewing insects on fruits and vegetables. Malathion is an effective insecticide for the control of several household pests such as houseflies, cockroaches, mosquitoes, aphids, animal ectoparasites, and human head and body lice. Malathion is also found in formulations with many other pesticides.
Verwenden
Malathion is used to control a wide spectrum of insect pests in many types of crops and disease vectors in public health programmes. It is also used to control ectoparasites of both humans and animals.
