| Gefahrenübersicht | EGME is toxic to the bone marrow and testicles. Workers exposed to high levels are at risk for granulocytopenia, macrocytic anemia, oligospermia, and azoospermia. A study of shipyard painters with evidence of testicular toxicity showed EGME TWA concentrations of up to 5.7 ppm (mean = 0.83 ppm) along with 2-ethoxyethanol TWA exposures at up to 22 ppm. Because EGME is readily absorbed through the skin, air levels are an inadequate assessment of exposure. In lethal concentration studies in rats, death is due to lung and kidney damage. Liver injury is observed in animals after subchronic inhalation studies. [ACGIH] CNS depression has not been seen after acute occupational exposure, but there have been reports of encephalopathy in workers exposed to 2-methoxyethanol over a period of weeks to months. [LaDou, p. 549] For ethylene glycol ethers, there is limited positive evidence of spontaneous abortions and decreased sperm counts in humans and strong positive evidence of birth defects and testicular damage in animals. [ATSDR Case Studies # 29] 53 workers exposed to EGME manufacturing copper-laminate circuit boards in Taiwan did not have evidence of liver toxicity compared to the control group. [Loh CH, et al. Hepatic Effects in Workers Exposed to 2-Methoxy Ethanol. J Occup Environ Med 200446:707-713] Potential adverse effects include injury to the bone marrow, liver, kidneys, and CNS (unconsciousness). May be a reproductive toxin, cause anemia, and defat the skin[ICSC] In animal experiments, causes congenital malformations and injury to the testesEvidence of reproductive effects in humans has not been convincing[REPROTOX] Labeled as May impair fertility and May cause harm to the unborn child by EU regulations[Glycol Ethers Online]See 2-Methoxyethanol. |

| Wechselwirkungen | ... A single exposure of mice to 125 ppm /2-methoxyethanol/ for 4 hr potentiated the hypnotic effects of barbituratesat 500 ppm for 4 hr, spontaneous motor activity decreased. The iv LD50 of serratia marcescens endotoxin in mice was 17 mg/kg. However, when the endotoxin was followed immediately by 950 mg iv methyl cellosolve, the LD50 dropped to 0.25 mg/kg. Ip methyl cellosolve also caused potentiation as did the pre-incubation of methyl cellosolve and endotoxin prior to injection. In adult female SPF Sprague-Dawley rats exposed for 2 hr to 1600 ppm 2-methoxyethanol (ME) the blood level of ME was considerably increased after pretreatment with ethanol (20 mmol/kg, ip). The blood level of ME remained nearly constant after the co-administration of ME (10 mm/kg, ip) with ethanol (20 mmol/kg), as long as ethanol levels in the blood remained above 3 mmol/l. Repeated ip dosing (five times one injection per hour) with ME (5mmol/kg) plus ethanol (8 or 10 mmol/kg) each resulted in an almost complete accumulation of ME. The prolonged retention of ME is due to an inhibition of its degradation via competition with ethanol for alcohol dehydrogenase.Ethylene glycol monomethyl ether (EGME) was administered to young male rats at doses varying from 50 to 500 mg/kg/day for 11 days. At sequential times animals were killed and testicular histology examined. The initial and major site of damage was restricted to the primary spermatocytes undergoing post zygotene meiotic maturation and division. When animals were treated with inhibitors of alcohol metabolism followed by a testicular toxic dose of EGME (500 mg/kg) an inhibitor of alcohol dehydrogenase (pyrazole) offered complete protection. However pretreatment with aldehyde dehydrogenase inhibitors disulfiram or pargyline did not ameliorate the testicular toxicity of EGME.For more Interactions (Complete) data for 2-METHOXYETHANOL (14 total), please visit the HSDB record page. |

| Umweltverhalten | TERRESTRIAL FATE: Based on a classification scheme(1), an estimated Koc value of 1(SRC), determined from a log Kow of -0.77(2) and a regression-derived equation(3), indicates that 2-methoxyethanol is expected to have very high mobility in soil(SRC). Volatilization of 2-methoxyethanol from moist soil surfaces is expected to be an environmental fate process(SRC) given a Henry's Law constant of 3.3X10-7 atm-cu m/mole(4). The potential for volatilization of 2-methoxyethanol from dry soil surfaces may exist(SRC) based upon a vapor pressure of 9.5 mm Hg(5). Utilizing the Japanese MITI test, 83% of the Theoretical BOD was reached in 2 weeks(6) indicating that biodegradation is an important environmental fate process in soil(SRC).AQUATIC FATE: Based on a classification scheme(1), an estimated Koc value of 1(SRC), determined from a log Kow of -0.77(2) and a regression-derived equation(3), indicates that 2-methoxyethanol is not expected to adsorb to suspended solids and sediment(SRC). Volatilization from water surfaces may occur(4) based upon a Henry's Law constant of 3.3X10-7 atm-cu m/mole(5). Using this Henry's Law constant and an estimation method(3), volatilization half-lives for a model river and model lake are 97 days and 700 days, respectively(SRC). According to a classification scheme(6), an estimated BCF of 3(SRC), from its log Kow(2) and a regression-derived equation(3), suggests the potential for bioconcentration in aquatic organisms is low(SRC). Utilizing the Japanese MITI test, 83% of the Theoretical BOD was reached in 2 weeks(7) indicating that biodegradation is an important environmental fate process in water(SRC).ATMOSPHERIC FATE: According to a model of gas/particle partitioning of semivolatile organic compounds in the atmosphere(1), 2-methoxyethanol, which has a vapor pressure of 9.5 mm Hg at 25 °C(2), is expected to exist solely as a vapor in the ambient atmosphere. Vapor-phase 2-methoxyethanol is degraded in the atmosphere by reaction with photochemically-produced hydroxyl radicals(SRC)the half-life for this reaction in air is estimated to be 31 hours(SRC), calculated from its rate constant of 1.25X10-11 cu cm/molecule-sec at 25 °C(3). 2-Methoxyethanol does not contain chromophores that absorb at wavelengths >290 nm(4) and, therefore, is not expected to be susceptible to direct photolysis by sunlight(SRC). |

| Toxizitätsübersicht | IDENTIFICATION AND USE: 2-Methoxyethanol (2-ME) is a colorless liquid with ether-like odor. It is used as a solvent for cellulose acetate, natural and synthetic resins, some alcohol-soluble dyesin nail polishes, quick-drying varnishes and enamels, and wood stains. It is also used as a as a jet fuel de-icer. HUMAN EXPOSURE AND TOXICITY: The substance is mildly irritating to the eyes and respiratory tract and may cause effects on the central nervous system, blood, bone marrow, kidneys and liver. Exposure at high levels could cause unconsciousness. The liquid defats the skin. Two non-fatal cases of poisoning by consuming 100 mL 2-ME in men aged 41 and 23 were reported. In addition to agitation and confusion, the predominant clinical features were nausea, cyanosis, hyperventilation, slight tachycardia, and metabolic acidosis. In one patient there were suggestions of moderate kidney failure. No evidence of liver damage was found and both patients recovered within 4 weeks. Chronic poisonings from vapors lead to toxic encephalopathy and evidence of bone marrow depression without hemolysis. Lung, kidney and liver changes have been observed. It was found that individuals exposed in utero to 2-ME showed characteristic dysmorphic features, unexplained mental retardation, and persistent cytogenetic damage. Exposure to 2-ME in utero could result in terminal chromosome rearrangements and shortening of telomere length. Epidemiological studies of workers exposed to 2-ME and 2-ethoxyethanol have shown some evidence of adverse effect on the male reproductive system, with an increased frequency of reduced sperm counts. ANIMAL TOXICITY STUDIES: In guinea pigs, 5 daily sc injections of either 0.5 or 1.0 mL/kg caused prostration, labored breathing, and death. Clinical exam and autopsy of the animals acutely treated with 2-ME revealed anuria, calcified casts in urine, irritation of bladder mucosa, hemorrhage in GI tract, lung edema, and liver and testicular injury. Evaluation of cryoprotectant toxicity showed that methanol and ME (5% and 10%) did not change the sperm motility after 30minMe(2)SO, DMA, and DMF (10% and 15%) and glycerol (5%, 10% and 15%) significantly decreased the motility of sperm within 1min after mixing. Based on these results, methanol and ME were selected as cryoprotectants (10%) to evaluate with different cooling rates (from 5 to 25 °C/min) and were compared to Me(2)SO and DMF (10%) (based on their use as cryoprotectants in previous publications). Post-thaw motility was affected by cryoprotectant, cooling rate, and their interaction (P</=0.000). The highest post-thaw motility (50+/-10%) was observed at a cooling rate of 10 °C/min with methanol as cryoprotectant. Comparable post-thaw motility (37+/-12%) was obtained at a cooling rate of 15 °C/min with ME as cryoprotectant. With DMF, post-thaw motility at all cooling rates was </=10% which was significantly lower than that of methanol and ME. With Me(2)SO, post-thaw motilities were less than 1% at all cooling rates, and significantly lower compared to the other three cryoprotectants (P</=0.000). When sperm from/AQUATIC SPECIES/ Growth inhibition of green algae by /2-methoxyethanol/ (2-ME) was noted at 104 mg/L and of cyanobacteria (blue-green algae) at 100 mg/L. |

| FIFRA-Anforderungen | As the federal pesticide law FIFRA directs, EPA is conducting a comprehensive review of older pesticides to consider their health and environmental effects and make decisions about their continued use. Under this pesticide reregistration program, EPA examines newer health and safety data for pesticide active ingredients initially registered before November 1, 1984, and determines whether the use of the pesticide does not pose unreasonable risk in accordance to newer saftey standards, such as those described in the Food Quality Protection Act of 1996. Pesticides for which EPA had not issued Registration Standards prior to the effective date of FIFRA '88 were divided into three lists based upon their potential for human exposure and other factors, with List B containing pesticides of greater concern than those on List C, and with List C containing pesticides of greater concern than those on List D. Methoxyethanol is found on List C. Case No: 3036Pesticide type: fungicide, antimicrobialCase Status: No products containing the pesticide are actively registered. Therefore, we are characterizing the case as "cancelled." Under FIFRA, pesticide producers may voluntarily cancel their registered products. EPA also may cancel pesticide registrations if registrants fail to pay required fees or make/meet certain reregistration commitments, or if EPA reaches findings of unreasonable adverse effects.Active ingredient (AI): methoxyethanolData Call-in (DCI) Date(s): 09/30/1992AI Status: The active ingredient is no longer contained in any registered products ... | Luftstandards | This action promulgates standards of performance for equipment leaks of Volatile Organic Compounds (VOC) in the Synthetic Organic Chemical Manufacturing Industry (SOCMI). The intended effect of these standards is to require all newly constructed, modified, and reconstructed SOCMI process units to use the best demonstrated system of continuous emission reduction for equipment leaks of VOC, considering costs, non air quality health and environmental impact and energy requirements. Ethylene glycol monomethyl ether is produced, as an intermediate or a final product, by process units covered under this subpart. |

Anwendungen & Verwendung
Beschreibung
2-Methoxyethanol (2-ME), also known as ethylene glycol monomethyl ether, is a glycol ether that has been known since the 1920s, but its use significantly increased in the 1970s. Methoxyethanol is an E-series glycol ether derived from methanol and ethylene oxide. Other commonly used glycol ethers include ethoxyethanol, butoxyethanol, and methoxypropanol.
Verwenden
2-Methoxyethanol is considered a non-comedogenic raw material. It is used as a solvent in nail products and as a stabilizer in cosmetic emulsions. It is able to penetrate the skin and may cause skin irritation.
