C(O[CH:4](OCC)[CH2:5][CH2:6][CH2:7][NH2:8])C.Cl.[Br:13][C:14]1[CH:15]=[C:16]([NH:21]N)[CH:17]=[CH:18][C:19]=1[F:20]>CO>[Br:13][C:14]1[CH:15]=[C:16]2[C:17]([C:5]([CH2:6][CH2:7][NH2:8])=[CH:4][NH:21]2)=[CH:18][C:19]=1[F:20]4,4-Diethoxybutan-1-amine (4.72 g, 29.3 mmol) was added to (3-bromo-4-fluorophenyl)hydrazine hydrochloride (6.4 g, 27 mmol). The resulting mixture in an open round bottom flask was placed into a preheated oil bath at 180° C. The mixture was stirred at 180° C. for 2.5 h and then cooled to 120° C. Methanol (300 mL) was added, and the mixture was stirred at ambient temperature for 18 h. The resulting suspension was filtered through a silica gel plug, and the silica gel was then washed with methanol (5×300 mL). The combined methanol fractions were concentrated under vacuum to provide the crude title compound (8.1 g) as a yellow solid, which was used in the next step without further purification: ESI MS m/z 257 [M+H]+.
