Methods of Manufacturing · 3 items

1,1,1,3,3,3-Hexafluoro-2-propanone is prepared from acetone in low yield by direct fluorination

1,1,1,3,3,3-Hexafluoro-2-propanone is manufactured by perchlorination of acetone, followed by reaction with hydrogen fluoride and a chromium catalyst. Separation of the product from excess hydrogen fluoride requires the use of a hydrogen fluoride acceptor, such as activated sodium fluoride or acetic anhydride.

The rearrangement of hexafluoropropyleneoxide induced by Lewis acids is an attractive new route that avoids the highly toxic sym-dichlorotetrafluoro- and chloropentafluoroacetones.

Preventive Measures · 1 item

SRP: The scientific literature for the use of contact lenses in industry is conflicting. The benefit or detrimental effects of wearing contact lenses depend not only upon the substance, but also on factors including the form of the substance, characteristics and duration of the exposure, the uses of other eye protection equipment, and the hygiene of the lenses. However, there may be individual substances whose irritating or corrosive properties are such that the wearing of contact lenses would be harmful to the eye. In those specific cases, contact lenses should not be worn. In any event, the usual eye protection equipment should be worn even when contact lenses are in place.

Environmental Fate · 3 items

TERRESTRIAL FATE: An estimated Koc of 150(1,SRC), based on an experimental log Kow(2), indicates that 1,1,1,3,3,3-hexafluoro-2-propanone will have moderate mobility in soil(3,SRC). In moist soils, 1,1,1,3,3,3-hexafluoro-2-propanone is expected to vigorously react with water producing 1,1,1,3,3,3-hexafluoro-2,2-propanediol, a stable solid hydrate as well as liquid hydrates(4,5). If released to dry soils, 1,1,1,3,3,3-hexafluoro-2-propanone may quickly volatilize based on a vapor pressure of 5 mm Hg at 25 °C(6,SRC). 1,1,1,3,3,3-Hexafluoro-2-propanone on soil surfaces may photolyse(SRC); this compound decomposes under UV irradiation to trifluoromethyl radicals and carbon monoxide(7).

AQUATIC FATE: 1,1,1,3,3,3-Hexafluoro-2-propanone is expected to vigorously react with water producing 1,1,1,3,3,3-hexafluoro-2,2-propanediol, a stable solid hydrate as well as liquid hydrates(1,2). This is expected to be a major fate process for 1,1,1,3,3,3-hexafluoro-2-propanone in aquatic systems(SRC). An estimated BCF value of 8(4), based on an experimental log Kow(3), indicates that 1,1,1,3,3,3-hexafluoro-2-propanone should not bioconcentrate in aquatic organisms(SRC).

ATMOSPHERIC FATE: Based on a vapor pressure of 5.0 mm Hg at 25 °C(1,SRC), as extrapolated from experimentally derived coefficients, 1,1,1,3,3,3-hexafluoro-2-propanone will exist solely in the vapor phase(2,SRC). 1,1,1,3,3,3-hexafluoro-2-propanone absorbs in the 220 to 400 nm region with a lambda max of 302 nm(2) and may thus directly photolyse(3,SRC). This compound decomposes under UV irradiation to trifluoromethyl radicals and carbon monoxide(3). The rate constant for the vapor-phase reaction of 1,1,1,3,3,3-hexafluoro-2-propanone with photochemically produced hydroxyl radicals has been estimated to be less than 1X10-16 cu cm/molecule-sec at 25 °C(4,SRC) indicating that 1,1,1,3,3,3-hexafluoro-2-propanone is essentially non-reactive and that degradation via hydroxyl radical reaction is not a major fate process for this compound in the atmosphere(SRC).

INHALATION EXPOSURES OF RATS INDICATED A 4-HR LETHAL CONCN OF 300 PPM; 200 PPM HFA (GAS) FOR 4 HR PRODUCED INJURY TO THE LIVER, KIDNEY, TESTES & THYMUS; & 100 PPM DAMAGED THE TESTES ONLY. DAILY, 6-HR REPEATED EXPOSURES OF RATS AT 60 PPM RESULTED IN DECREASED SPERMATOGENESIS AFTER 10 EXPOSURES & DECREASED BONE-MARROW ERYTHROPOIESIS AFTER TERMINATION OF EXPOSURE. TESTICULAR REGENERATION WAS STILL INCOMPLETE AT 57 DAYS FOLLOWING A SINGLE, 4-HR EXPOSURE AT 60 PPM HFA.

NINETY DAYS EXPOSURE AT 0.1 PPM RESULTED IN NO GROSS BIOCHEMICAL, HEMATOLOGIC OR HISTOPATHOLOGIC CHANGES IN EITHER RATS OR DOGS, ALTHOUGH THE LUNG WEIGHTS OF THE DOGS EXCEEDED THAT OF THE CONTROLS. AT 1 PPM, REVERSIBLE RENAL DYSFUNCTION IN RATS & INCREASED LUNG WEIGHT IN DOGS OVER THAT OF CONTROLS WAS ALL THAT WAS NOTED OF SIGNIFICANCE, NO HISTOPATHOLOGIC CHANGES. AT 12 PPM, SERIOUS TISSUE DAMAGE OCCURRED; THE RATE OF WEIGHT GAIN, ADRENAL & TESTES WEIGHTS OF MALE RATS, WERE LESS THAN THOSE OF CONTROLS. TRENDS TOWARD INCREASED WEIGHTS OF HEART, STOMACH, LIVER & LUNG WERE OBSERVED. IN ADDITION, THERE WAS EVIDENCE OF A TREND TOWARD LYMPHOCYTOSIS & RENAL DYSFUNCTION. HISTOPATHOLOGICALLY, SEVERE, BUT REVERSIBLE, TESTICULAR DAMAGE, & SLIGHT HYPERPLASIA OF THE SPLEEN, THYMUS, LYMPH NODES & BONE MARROW OCCURRED.

... SEVERE INJURY FROM 12 PPM LEVEL /HAS/ RESULTED IN DOGS; THE TESTES WEIGHT DECREASED, & THE WEIGHTS OF PITUITARY & LUNG INCREASED. DOGS ALSO DEVELOPED NEUTROPHILIA, HYPERGLYCEMIA, HYPOALBUMINERIA, & ANEMIA. AS WITH RATS, THE DOGS DEVELOPED SEVERE, BUT REVERSIBLE, TESTICULAR DAMAGE & SLIGHT HYPOPLASIA OF THE SPLEEN, THYMUS & LYMPH NODES. /FROM 90 DAYS INHALATION/

Testicular degeneration and atrophy were seen in rats given dermal doses of 65, 130, or 260 mg/kg 5 days/wk for 2 wk. These changes were still apparent 14 days after the last dose. They were not produced by a dermal dosage of 13 mg/kg for 14 days. The dose of 13 mg/kg/day HFA was considered a no-observed-effect level (NOEL). The structural changes (testicular atrophy) and altered lipid metabolism correlated well. In these studies, changes in steroidogenesis was due to inhibition in Leydig cells and was not due to a direct or immediate effect of HFA nor by hormonal mediation. HFA (as the sesquihydrate) was applied to the skin of pregnant rats at various dosages on days 6 through 16 of gestation. Maternal toxicity was observed at 90 mg/kg/day. The number of resorptions and live fetuses per litter suggested fetal toxicity at 25 mg/kg with fetal size being reduced at the 5 and 25 mg/kg doses (but not at 1 mg/kg). A small increase in soft tissue and external abnormalities was seen, but a definite teratogenic response could not be concluded. /Sesquihydrate/

General DescriptionHexafluoroacetone is a colorless, toxic, and highly reactive gas. At ambient temperatures, Hexafluoroacetone is likely to generate a considerable amount of vapor. Hexafluoroacetone is an irritant to skin, eyes and mucous membranes and is toxic by ingestion, skin absorption, and inhalation. When heated to high temperatures Hexafluoroacetone emits toxic fluoride fumes. Prolonged exposure of the container to fire or intense heat may cause Hexafluoroacetone to violently rupture and rocket. Hexafluoroacetone is used in the production of other chemicals.
