Hazards Summary · 2 items

For the triazine herbicides, systemic toxicity unlikely unless large amounts are ingested; May cause irritation; [EPA Pesticides, p. 121] A mild eye irritant; [HSDB] May cause irritation; [MSDSonline] 2021 TLV Basis: liver and kidney damage, bone marrow effects. and maternal and fetal toxicity; No human studies available; [ACGIH]

The major hazards encountered in the use and handling of prometryne stem from its toxicologic properties. Toxic by inhalation and ingestion, exposure to this colorless to white, crystalline substance may occur from its manufacture, formulation, and use as an herbicide. Effects from exposure may include shortness of breath, muscle spasms, ataxia, and anorexia. In activities and situations where over exposure may occur, wear a self-contained breathing apparatus and personal protective clothing. If contact should occur, immediately flush affected skin or eyes with running water for at least 15 minutes. Remove contaminated clothing and shoes at the site. While prometryne does not ignite easily, it may burn with the production of irritating and poisonous gases. For fires involving prometryne, extinguish with dry chemical, CO2, Halon, water spray, fog, or standard foam. Prometryne may be shipped domestically via air, rail, road, and water, in containers bearing the label "Poison". Small dry spills of prometryne may be placed into a clean, dry, covered container for later disposal (liquid solutions are first absorbed in sand or other noncombustible absorbent). Large liquid spills should be diked far ahead to prevent prometryne from entering water sources and sewers. Before implementing land disposal of prometryne, consult with regulatory agencies for guidance.

Environmental Fate · 3 items

TERRESTRIAL FATE: Based on a classification scheme(1), Koc values of 39.4 to 3,473(2), indicate that prometryne is expected to have very high to slight mobility in soil(SRC). Volatilization of prometryne from moist soil surfaces is not expected to be an important fate process(SRC) given an estimated Henry's Law constant of 1.2X10-8 atm-cu m/mole(SRC), derived from its vapor pressure, 1.24X10-6 mm Hg(3), and water solubility, 33 mg/L(4). In a model terrestrial system, no volatilization of the parent compound was seen from soil surfaces(7). However, estimated half-lives for volatilization of prometryne from soil range from 41 to 60 days for shallow to deep placement and varying water evaporation rates(5). The loss of prometryne applied to the surface of soil has also been observed to be relatively rapid in the first 30-40 days compared to the loss of prometryne which was incorporated into the soil(6). Prometryne is not expected to volatilize from dry soil surfaces(SRC) based upon its vapor pressure(3). Half-lives for the biodegradation of prometryne in aerobic and anaerobic soil have been reported as 150 and 360 days, respectively(8).

AQUATIC FATE: Based on a classification scheme(1), Koc values of 39.4 to 3,473(2), indicate that prometryne may adsorb to suspended solids and sediment(SRC). Volatilization from water surfaces is not expected(3) based upon an estimated Henry's Law constant of 1.2X10-8 atm-cu m/mole(SRC), derived from its vapor pressure, 1.24X10-6 mm Hg(4), and water solubility, 33 mg/L(5). According to a classification scheme(6), an estimated BCF of 48(SRC), from a log Kow of 3.5(7) and a regression-derived equation(8), suggests the potential for bioconcentration in aquatic organisms is moderate(SRC). Half-lives for the biodegradation of prometryne in aerobic and anaerobic soil have been reported as 150 and 360 days, respectively(8), suggesting that biodegradation is not an important environmental fate process in water(SRC). Photochemical degradation half-lives of prometryne have a range of 4.6 hours(9) to 88 days (10).

ATMOSPHERIC FATE: According to a model of gas/particle partitioning of semivolatile organic compounds in the atmosphere(1), prometryne, which has a vapor pressure of 1.24X10-6 mm Hg at 25 °C(2), will exist in both the vapor and particulate phases in the ambient atmosphere. Vapor-phase prometryne is degraded in the atmosphere by reaction with photochemically-produced hydroxyl radicals(SRC); the half-life for this reaction in air is estimated to be 10 hours(SRC), calculated from its rate constant of 3.8X10-11 cu cm/molecule-sec at 25 °C(SRC) that was derived using a structure estimation method(3). Particulate-phase prometryne may be removed from the air by wet or dry deposition(SRC). Prometryne does contain chromophores that absorb at wavelengths >290 nm and therefore may be susceptible to direct photolysis by sunlight(4).

Food Survey ValuesPrometryne was not detected (detection limit 10 ppb) in annual composite samples of Ontario-grown potatoes analyzed from 1983 to 1985(1).
Adverse EffectsOccupational hepatotoxin - Secondary hepatotoxins: the potential for toxic effect in the occupational setting is based on cases of poisoning by human ingestion or animal experimentation.

FIFRA Requirements · 4 items

Tolerances are established for residues of the herbicide prometryn (2,4-bis(isopropylamino)-6-methylthio-s-triazine) in or on the following raw agricultural commodities: [Table#4780]

Tolerances with regional registration, as defined in §180.1(n), are established for residues of the herbicide prometryn (2,4-bis(isopropylamino-6-methylthio-s- triazine) in or on the following raw agricultural commodity: [Table#4781]

Section 4(g)(2)(A) of FIFRA calls for the Agency to determine, after submission of relevant data concerning an active ingredient, whether products containing the active ingredient are eligible for reregistration. The Agency has previously identified and required the submission of the generic (i.e. active ingredient specific) data required to support reregistration of products containing prometryn. The Agency has completed its review of these generic data, and has determined that the data are sufficient to support reregistration of all products containing prometryn.

As the federal pesticide law FIFRA directs, EPA is conducting a comprehensive review of older pesticides to consider their health and environmental effects and make decisions about their future use. Under this pesticide reregistration program, EPA examines health and safety data for pesticide active ingredients initially registered before November 1, 1984, and determines whether they are eligible for reregistration. In addition, all pesticides must meet the new safety standard of the Food Quality Protection Act of 1996. Prometryn is found on List A, which contains most food use pesticides and consists of the 194 chemical cases (or 350 individual active ingredients) for which EPA issued registration standards prior to FIFRA '88. Case No: 0467; Pesticide type: herbicide; Registration Standard Date: 03/20/87 PB87-184826; Case Status: RED Approved 09/95; OPP has made a decision that some/all uses of the pesticide are eligible for reregistration, as reflected in a Reregistration Eligibility Decision (RED) document ; Active ingredient (AI): prometryn; Data Call-in (DCI) Date(s): 09/30/91, 08/15,96; AI Status: OPP has completed a Reregistration Eligibility Decision (RED) for the case/AI.

In addition to CH3S- displacement by -OH, in rats and rabbits treated with prometryne CH3S-group is dealkylated without sulfur oxidation followed by oxidative coupling to disulfide.

... Hydroxypropazine & hydroxyatrazine were detected when cotton, soybeans, peanuts, & carrots were treated with (14)C-ring-labeled prometryn ... Supporting evidence for methylthio group cleavage came from studies with (14)CH3-labeled prometryn in peas where moderate amount of (14)carbon dioxide were liberated.

Rats & rabbits fed prometryne excreted 2-mercapto-4-amino-6-isopropylamino-s-triazine & bis(2-amino-4-isopropylamino-s-triazinyl-6-yl)disulfide. Mono- & di-dealkylated prometryne analogs were also observed. 8 day old pea plants were exposed ... & harvested 8 days later. Several metab pathways were indicated. In one, the side chain of prometryn was metab and 2-hydroxy-4-amino-6-isopropylamino-s-triazine was identified. In the other, the methylthio group was oxidized to sulfone which readily hydrolyzes to hydroxy analog. Chromatography indicated presence of sulfoxide & hydroxy analogs.

Prometryn in rats and rabbits is converted to both the thio ... /deriv/, & the corresponding disulfide, together with a number of other urinary metabolites.

Mono- & didealkylated parent compounds ... were identified in urine of rats & rabbits after application of several hundred mg/kg of nonlabeled simazine, atrazine, propazine, prometone and prometryn.

In the rat, most of the administered prometryn is excreted within 48 hours in urine and feces.

When given by oral gavage to rats as a (14)C-labeled material, 2,4-bis(isopropylamino)-6-methylthio-s-triazine was detected in both urine and feces. The chemical was absorbed and rapidly excreted. Limited transfer to the milk of lactating goats or to the eggs of laying hens was reported. The triazines, including prometryne, are generally well-absorbed by the mammalian gut and probably across the skin. The breakdown of prometryne is not adequately understood ... No detectable residues of prometryne or its metabolites were found in the muscle, fat, blood, liver, kidney, or other organs of sheep and cattle fed up to 100 ppm for 4 weeks. However, prometryne or its breakdown products were found in whole milk samples taken from cows that were fed up to 100 ppm in their diets for 21 days.

They are efficiently absorbed from intestine, and presumably there is some absorption across the skin and lung. /Urea-, uracil- and triazine-based herbicides/
