Hazards Summary · 2 件

A strong skin, eye, and mucous membrane irritant; [HSDB] May burn skin and eyes; [CAMEO] See Arsenic and linked occupational diseases.

The major hazards encountered in the use and handling of phenyldichloroarsine stem from its toxicologic properties. Toxic by all routes (ie, inhalation, ingestion, and dermal contact), exposure to this liquid material may occur from its manufacture or use as a lacrymator, and solvent for diphenylcyanoarsine. Effects from exposure may include contact burns to the skin and eyes, he adache, generalized weakness, nausea, hemoglobinuria, and death (if sufficient quantities are inhaled). OSHA has established a time-weighted average (TWA) limit of 0.5 mg/cu m for organic arsenic compounds. Mechanical ventilation should be used to maintain phenyldichloroarsine at, or below the OSHA limits. In activities and situations where over-exposure may occur, wear a positive pressure, self-contained breathing apparatus and chemical protective clothing which is specifically recommended by the shipper or manufacturer. If exposure should occur, immediately flush exposed eyes and skin with running water for at least 15 minutes. Remove contaminated clothing at the site. While phenyldichloroarsine does not ignite easily, it can burn with the production of poisonous gases. Also, containers of this substance may explode violently in the heat of a fire. For fires involving phenyldichloroarsine, extinguish with dry chemical, CO2, water spray, fog, or standard foam. Dike fire control water and do not scatter the material. Phenyldichloroarsine may be shipped domestically via air, rail, road, and water, in containers bearing the label, "Poison." For small spills of this substance, take up with sand or other noncombustible absorbent and place into containers for later disposal. Dike far ahead of large spills for later disposal.

Mechanism of Action · 2 件

In order to investigate the mechanism of cytotoxicity of organic arsenicals toward lipoic acid containing enzymes, a lipoic acid-phenyldichlorarsine adduct was prepared in methanol, and the structure and molecular motions were studied. A six-membered heteroatom adduct was formed. One dimensional and 2 dimensional NMR spectroscopy was used to confirm the structure. Spin-lattice relaxation times of the various carbon atoms indicated that the overall molecular reorientation time of the molecule is 0.02 nsec at 30 °C. An Arrhenius plot of the data showed that the activation energy for molecular tumbling is 13.4 kJ/mol. The system did not demonstrate dimerization as the bulky side chain of lipoic acid prevented dimerization from occurring.

There remains considerable controversy regarding the mode of action of the phenylarsonic compounds. However, they seem to have an action different from that of inorganic, aliphatic, and aromatic trivalent arsenic compounds. The arsenic incorporated in the feed additives is in the pentavalent form, and it is likely that the phenylarsonic compounds have their primary action as pentavalent arsenicals; this may account for their distinctive effects in birds and animals. Some workers have suggested that both the toxicity and the efficiency of these compounds are due to their degradation and reduction to inorganic trivalent forms. Others have clearly established that these compounds are excreted unchanged by chickens and that there is no evidence that they are converted to inorganic arsenic. Similar experiments by other workers with rats, rabbits, and swine indicate that the phenylarsonic compounds are for the most part excreted unchanged by the kidneys, although some apparently undergo a limited biotransformation. /Phenylarsonic compounds/

Metabolism/Metabolites · 1 件

Arsenic is absorbed mainly by inhalation or ingestion, as to a lesser extent, dermal exposure. It is then distributed throughout the body, where it is reduced into arsenite if necessary, then methylated into monomethylarsenic (MMA) and dimethylarsenic acid (DMA) by arsenite methyltransferase. Arsenic and its metabolites are primarily excreted in the urine. Arsenic is known to induce the metal-binding protein metallothionein, which decreases the toxic effects of arsenic and other metals by binding them and making them biologically inactive, as well as acting as an antioxidant. SRP: Contaminated protective clothing should be segregated in such a manner so that there is no direct personal contact by personnel who handle, dispose, or clean the clothing. Quality assurance to ascertain the completeness of the cleaning procedures should be implemented before the decontaminated protective clothing is returned for reuse by the workers. Contaminated clothing should not be taken home at end of shift, but should remain at employee's place of work for cleaning.

SRP: The scientific literature for the use of contact lenses in industry is conflicting. The benefit or detrimental effects of wearing contact lenses depend not only upon the substance, but also on factors including the form of the substance, characteristics and duration of the exposure, the uses of other eye protection equipment, and the hygiene of the lenses. However, there may be individual substances whose irritating or corrosive properties are such that the wearing of contact lenses would be harmful to the eye. In those specific cases, contact lenses should not be worn. In any event, the usual eye protection equipment should be worn even when contact lenses are in place.

Environmental Fate · 3 件

TERRESTRIAL FATE: Based on a classification scheme(1), an estimated Koc value of 820(SRC), determined from a structure estimation method(2), indicates that phenyldichloroarsine is expected to have moderate mobility in soil(SRC). Volatilization of phenyldichloroarsine from moist soil surfaces is expected to be an important fate process(SRC) given an estimated Henry's Law constant of 3.0X10-5 atm-cu m/mole(SRC), using a fragment constant estimation method(3). However, phenyldichloroarsine rapidly hydrolyzes in water(4); therefore, volatilization may be less important than estimated(SRC). Phenyldichloroarsine is not expected to volatilize from dry soil surfaces(SRC) based upon a vapor pressure of 1.13X10-1 mm Hg(5).

AQUATIC FATE: Based on a classification scheme(1), an estimated Koc value of 820(SRC), determined from a structure estimation method(2), indicates that phenyldichloroarsine is expected to adsorb to suspended solids and sediment(SRC). Volatilization from water surfaces is expected(3) based upon an estimated Henry's Law constant of 3.0X10-5 atm-cu m/mole(SRC), developed using a fragment constant estimation method(4). Using this Henry's Law constant and an estimation method(3), volatilization half-lives for a model river and model lake are 31 hours and 19 days, respectively(SRC). According to a classification scheme(5), an estimated BCF of 46(SRC), from an estimated log Kow of 3.1(2) and a regression-derived equation(6), suggests the potential for bioconcentration in aquatic organisms is low(SRC). However, phenyldichloroarsine rapidly hydrolyzes in water(7); therefore, volatilization and bioconcentration may be less important than estimated(SRC).

ATMOSPHERIC FATE: According to a model of gas/particle partitioning of semivolatile organic compounds in the atmosphere(1), phenyldichloroarsine, which has a vapor pressure of 1.13X10-1 mm Hg at 25 °C(2), is expected to exist solely as a vapor in the ambient atmosphere(SRC). Vapor-phase phenyldichloroarsine is degraded in the atmosphere by reaction with photochemically-produced hydroxyl radicals(SRC); the half-life for this reaction in air is estimated to be 8 days(SRC), calculated from its rate constant of 2.0X10-12 cu cm/molecule-sec at 25 °C(SRC) determined using a structure estimation method(3).

Toxicity Summary · 1 件

Arsenic and its metabolites disrupt ATP production through several mechanisms. At the level of the citric acid cycle, arsenic inhibits pyruvate dehydrogenase and by competing with phosphate it uncouples oxidative phosphorylation, thus inhibiting energy-linked reduction of NAD+, mitochondrial respiration, and ATP synthesis. Hydrogen peroxide production is also increased, which might form reactive oxygen species and oxidative stress. Arsenic's carginogenicity is influenced by the arsenical binding of tubulin, which results in aneuploidy, polyploidy and mitotic arrests. The binding of other arsenic protein targets may also cause altered DNA repair enzyme activity, altered DNA methylation patterns and cell proliferation. (T1, A17)

The purpose of the study was to identify binding sites of organic arsenic in the erythrocyte and to explain species differences in binding. Washed erythrocytes were exposed to graded concentrations of (14)C-phenyldichloroarsine (PDA) in phosphate-buffered saline containing 0.1% glucose and 0.1% bovine serum albumin. At low phenyldichloroarsine concentrations, all cells bind the arsenical rapidly (within 10 min) and quantitatively. Human, pig, hamster, guinea pig, and mouse erythrocytes approached saturation at 0.02-0.3 mumol phenyldichloroarsine/10(9) cells, depending on the species. Saturation points correlated well with each respective species erythrocyte glutathione content. In contrast, rat erythrocytes showed no sign of saturation at phenyldichloroarsine loads as high as 3.0 mumol/10(9) cells. Hemolysates of phenyldichloroarsine-treated erythrocytes were subjected to Sephadex G-75 gel filtration chromatography. (14)C from rat hemolysate was distributed between the hemoglobin and small molecular weight (glutathione-containing) fractions. In all other species, the (14)C eluted almost exclusively with the glutathione-containing fractions. In equilibrium dialysis experiments, human hemoglobin did not bind phenyldichloroarsine, whereas rat hemoglobin bound 2 phenyldichloroarsine/mol with Kd approximately 5 m. In conclusion, glutathione is the principle binding site of phenyldichloroarsine in erythrocytes. In most species, the arsenical does not bind to hemoglobin, even though it has free (titratable) sulfhydryls considerably in excess of the glutathione concentration. In rat erythrocytes, phenyldichloroarsine binds both to glutathione and to hemoglobin.
