Emergency treatment: Plants- pyrrolizidine alkaloids; Pyrrolizidine alkaloids (PAs) cause veno-occlusive liver disease. Diagnosis is difficult because symptoms may be delayed for days or weeks. After drinking herbal tea or eating plants contaminated with seeds of PA plants, symptoms may include vomiting, abdominal pain, hepatomegaly, and later cirrhosis and ascites. Jaundice may be present. Children may appear to have Reye's syndrome. [HSDB] Teas containing PAs include coltsfoot, comfrey, gordolobo, groundsel, mate, tansy ragwort, and T'u-san-chi. The teas may be prescribed as herbal remedies for anxiety or amenorrhea. The alkaloid is concentrated in the root of the plants. As little as 85 mg may be toxic. [Haddad, p. 1082]

Hepatotoxic pyrrolizidine alkaloid found in Heliotropium lasiocarpum; [Merck Index] Also found in other plants of the Boraginaceae family; A known liver carcinogen with antimitotic activity used for research; [HSDB] More than 350 pyrrolizidine alkaloids (PAs) in over 6000 plants have been identified. They are probably the most common cause of plant poisoning in the world and affect livestock, wildlife, and humans. They are found in the Boraginaceae, Compositae, and Leguminosae families and are commonly encountered in the following plants: fiddle neck, tarweed, rattle box, blue weed, giant hog weed, heliotrope, groundsels, Senecio, and comfrey. [Haddad, p. 494] Liver disease from bush tea derived from the Crotalaria plant is endemic in Jamaica. Cereal contaminated with PA seeds (Heliotropium and Crotalaria) have caused epidemics in Afghanistan and India. Herbal remedies that have caused liver disease include Senecio and comfrey. LASIOCARPINE (7.8 MG/KG, IP TWICE WEEKLY FOR 4 WK, THEN ONCE A WK FOR 18 WK) DID NOT PREVENT INITIATION OF LIVER TUMORS BY AFLATOXIN B1 (2 PPM IN DIET), BUT ALTERED THE PATHOGENIC PATTERN IN THAT, IN RATS RECEIVING AFLATOXIN & LASIOCARPINE, THE LIVER TUMORS WERE ASSOC WITH POSTNECROTIC CIRRHOSIS, WHEREAS IN RATS DEVELOPING LIVER TUMORS WITH AFLATOXIN ALONE THEY WERE NOT.

SENSITIVITY OF RATS TO LASIOCARPINE IS REDUCED BY FEEDING A DIET LOW IN LIPOTROPES OR BY SIMULTANEOUS ADMIN OF MERCAPTOETHYLAMINE.

PREGNENOLONE-16ALPHA-CARBONITRILE, SPIRONOLACTONE, ETHYLESTRENOL, PHENOBARBITAL, & DIPHENYLHYDANTOIN PREVENTED THE MORTALITY & LIVER DAMAGE CAUSED BY SINGLE DOSE OF LASIOCARPINE (100 MG/KG, IP) WHEREAS SKF 525-A, OR PARTIAL HEPATECTOMY, GREATLY INCR LETHALITY.

RATS WERE TREATED CONCURRENTLY WITH THIOACETAMIDE & THE PYRROLIZIDINE ALKALOID LASIOCARPINE. THIOACETAMIDE WAS GIVEN IP IN DOSE OF 50 MG/KG BODY WT TWICE WEEKLY & LASIOCARPINE WAS ADMIN IN THE DIET AT CONCN OF 50 PPM. THIOACETAMIDE & LASIOCARPINE PRODUCED NUMEROUS GROSSLY VISIBLE GREY NODULES IN LIVERS OF 26 OF 30 RATS. Toxicity Summary · 1 件

Lasiocarpine is toxic to liver cells but not to lung cells, which is unable to metabolize the pyrrolizidine alkaloids to pyrroles. The application of lasiocarpine to human liver cells in culture is followed by inhibition of DNA, RNA and protein synthesis; vacuolation of the cells, the prevention of mitosis and the formation of giant cells (“megalocytes”). (A15431) Effects on the liver include hemorrhagic necrosis of parenchymal cells, marked enlargement of parenchymal cells and of their nuclei and nucleoli, fibrosis or cirrhosis, depression of cell division, and possibly neoplasia. (A15432) The inducibility of tryptophan pyrrolase activity by hydrocortisone, in the livers of rats treated chronically with lasiocarpine is an indication that translational mechanisms are intact. However, the increased uptake of 3H-thymidine by megalocytes, in the absence of observable mitotic activity, suggests that these cells are in the process of hypertrophy. They may act as prolonged, potent inhibitors of mitosis in hepatocytes, they are mutagenic in Drosophila and cause chromosome breakage in leukocytes. One of the unusual results of chronic poisoning by lasiocarpine and other members of the pyrrolizidine alkaloids is progressive enlargement of liver cells to up to three times the normal diameter, with an accompanying proportionate increase in nuclear diameter. The term megalocytosis has been applied to this abnormality which may be associated with mild fibrosis of central veins and proliferation of bile ducts. The prominence of rough ER in enlarged hepatocytes is s a sign of increased metabolic activity. (A15433)

A BIOASSAY OF LASIOCARPINE FOR POSSIBLE CARCINOGENICITY WAS CONDUCTED BY ADMIN THE TEST CHEM IN THE DIET TO FISCHER 344 RATS. GROUPS OF 24 RATS OF EACH SEX WERE ADMIN LASIOCARPINE AT 7, 15, OR 30 PPM, FOR 104 WK. MATCHED CONTROLS CONSISTED OF GROUPS OF 24 UNTREATED RATS OF EACH SEX. THERE WAS A POS DOSE-RELATED TREND IN THE INCIDENCE OF ANGIOSARCOMA OF THE LIVER. METASTATIC ANGIOSARCOMAS WERE PRESENT IN THE LUNGS FROM A FEW RATS IN ALL 3 TREATED GROUPS OF BOTH SEXES. THUS, UNDER THE CONDITIONS OF THIS BIOASSAY LASIOCARPINE WAS CARCINOGENIC FOR LIVER OF FISCHER 344 RATS.

RESEARCH CHEMICAL

KNOWN LIVER CARCINOGEN WITH ANTIMITOTIC ACTIVITY USED IN RESEARCH

Lasiocarpine is a pyrrolizidine alkaloid that is found in the seeds of Heliotropium lasiocarpum, Heliotropium europaeum, and several other plant species, all members of the family Boraginaceae.

Methods of ManufacturingLASIOCARPINE IS PRODUCED FOR RESEARCH PURPOSES FROM HELIOTROPIUM EUROPAEUM BY TWO FIRMS IN AUSTRALIA.
General Manufacturing Information · 2 件

THE PRESENCE OF SODIUM CHLORIDE & AMMONIUM CHLORIDE REDUCED THE EXTRACTION OF LASIOCARPINE WITH CHLOROFORM FROM ALKALINE SOLN. THE EXTRACTION OF LASIOCARPINE FROM ACIDIC SOLN DEPENDED ON THE ALKALOID CONCN & THE TYPE OF ALKALOID. IN THE PRESENCE OF AMMONIUM SULFATE, AT PH 2.5, NO ALKALOID WAS EXTRACTED, BUT IN THE PRESENCE OF SODIUM CHLORIDE THE EXTRACTION WAS 32-68%. FROM ACID OR ALKALINE SOLN WITH ETHER OR BENZENE, EXTRACTION WAS NOT SIGNIFICANTLY AFFECTED BY ELECTROLYTES.

HELIOTROPIUM EUROPAEUM L IS REPORTED TO HAVE BEEN USED MEDICINALLY IN INDIA AS AN EMETIC & IN THE TREATMENT OF SNAKE BITES. IT HAS ALSO BEEN USED MEDICINALLY IN GREECE & THE EASTERN MEDITERRANEAN.

説明A hepatotoxic alkaloid found in Heliotropiurn lasiocarpurn, the base forms colourless leaflets from petroleum ether and has [α]20D - 4° (EtOH). It is readily soluble in C6H6 or EtOH but only slightly so in H20. On alkaline hydrolysis it furnishes angelic acid and heliotridine, C8H1302N, m.p. 116.5-118°C, forming a hydrochloride, m.p. 122-4°C and the benzoyl derivative hydrochloride, m.p. 180°C. On catalytic hydrogenation it yields a base, C13H230 2N, b.p. 123-5°Cj 8 mm; [α]D + 3.8° (EtOH), yielding a crystalline picrate, m.p. 157 _9°C, and lasiocarpic acid, C8H160 S, m.p. 95-7°C; [α]D + 10.6° (EtOH). The structure is therefore angelyllasiocarpylheliotridine, both hydroxyls in heliotridine being es terifie d
