TERRESTRIAL FATE: Based on a classification scheme(1), an estimated Koc value of 880(SRC), determined from a structure estimation method(2), indicates that p,p'-oxybis(benzenesulfonyl hydrazide) is expected to have low mobility in soil(SRC). Volatilization of p,p'-oxybis(benzenesulfonyl hydrazide) from moist soil surfaces is not expected to be an important fate process(SRC) given an estimated Henry's Law constant of 1.26X10-17 atm-cu m/mole(SRC), using a fragment constant estimation method(3). p,p'-Oxybis(benzenesulfonyl hydrazide) is not expected to volatilize from dry soil surfaces(SRC) based upon an estimated vapor pressure of 6.91X10-11 mm Hg at 25 °C(SRC), determined from a fragment constant method(4). A 2% of theoretical BOD using activated sludge in the Japanese MITI test(5) suggests that biodegradation is not an important environmental fate process in soil(SRC).
AQUATIC FATE: Based on a classification scheme(1), an estimated Koc value of 880(SRC), determined from a structure estimation method(2), indicates that p,p'-oxybis(benzenesulfonyl hydrazide) is expected to adsorb to suspended solids and sediment(SRC). Volatilization from water surfaces is not expected(3) based upon an estimated Henry's Law constant of 1.26X10-17 atm-cu m/mole(SRC), developed using a fragment constant estimation method(4). According to a classification scheme(5), BCF values of less than 3.0(6), suggest bioconcentration in aquatic organisms is low(SRC). A 2% of theoretical BOD using activated sludge in the Japanese MITI test(6) suggests that biodegradation is not an important environmental fate process in water(SRC).
ATMOSPHERIC FATE: According to a model of gas/particle partitioning of semivolatile organic compounds in the atmosphere(1), p,p'-oxybis(benzenesulfonyl hydrazide), which has an estimated vapor pressure of 6.91X10-11 mm Hg at 25 °C(SRC), determined from a fragment constant method(2), is expected to exist solely in the particulate phase in the ambient atmosphere. Particulate-phase p,p'-oxybis(benzenesulfonyl hydrazide) may be removed from the air by wet or dry deposition(SRC). p,p'-Oxybis(benzenesulfonyl hydrazide) does not contain chromophores that absorb at wavelengths >290 nm(3), and therefore is not expected to be susceptible to direct photolysis by sunlight(SRC).

/GENOTOXICITY/ The test substance was evaluated for its ability to induce chromosomal aberrations in primary human lymphocyte cells in the presence and absence of a rat liver homogenate metabolic activation system. The test article was tested at the following concentrations: Neat and 1:2, 1:4, 1:8, 1:16, 1:32, and 1:64 dilutions of the neat extract. The concentrations chosen to be scored for the activated assay were neat and 1:2 and 1:4 dilutions of the test extract. The results indicate that the test substance did not cause a statistically significant increase in the number of chromosome aberrations (p < 0.05). This was observed for both activated and non-activated system. In addition, there was no detectable dose response in the number of aberrations of both activated and non-activated systems, verifying the validy of the test system. In conclusion, the test substance did not induce aberrations in primary cultured human lymphocytes and is considered non-clastogenic.
AEROBIC: p,p'-Oxybis(benzenesulfonyl hydrazide), present at 100 ppm, reached 2% of its theoretical BOD in 2 weeks using an activated sludge inoculum at 30 ppm in the Japanese MITI test(1,2), indicating that biodegradation is not expected to be an important environmental fate process(SRC).
BCF values of <0.3 and <3.0 were calculated in carp (Cyprinus carpio) for p,p'-oxybis(benzenesulfonyl hydrazide), exposed over a 6 week test period to 1 mg/L and 0.1 mg/L, respectively(1). According to a classification scheme(2), this BCF suggests the potential for bioconcentration in aquatic organisms is low(SRC).
The Henry's Law constant for p,p'-oxybis(benzenesulfonyl hydrazide) is estimated as 1.26X10-17 atm-cu m/mole(SRC) using a fragment constant estimation method(1). This Henry's Law constant indicates that p,p'-oxybis(benzenesulfonyl hydrazide) is expected to be essentially nonvolatile from water surfaces(2). p,p'-Oxybis(benzenesulfonyl hydrazide) is not expected to volatilize from dry soil surfaces(SRC) based upon an estimated vapor pressure of 6.91X10-11 mm Hg(SRC), determined from a fragment constant method(3).
/LABORATORY ANIMALS: Acute Exposure/ There is limited evidence that 4,4'-oxybis(benzenesulphonyl hydrazide) irritated the skin and eyes of laboratory animals. A moderate to low acute oral toxicity was seen in rats and mice; effects on the nervous system were observed.
/LABORATORY ANIMALS: Acute Exposure/ Following repeated oral exposure, liver, kidney and adrenal effects were seen in limited studies in rats.
/LABORATORY ANIMALS: Acute Exposure/ Mutagenic activity was demonstrated in bacterial assays, including an Ames test, and DNA damage was induced in cultures of rat and mouse cells. No evidence of genotoxic potential was found in mice treated by intraperitoneal administration.
/GENOTOXICITY/ Bacterial reverse mutation assay. Benzenesulfonic acid, 4, 4'-oxybis-, dihydrazide was mutagenic in Salmonella typhimurium (Ames test) in the presence or absence of a liver metabolic activation system. In a test with one strain of Escherichia coli, a liver metabolic activation system was required for the hydrazide to exhibit mutagenic potential, although tests with other strains found no evidence of mutagenicity in the presence or absence of a liver metabolic fraction.
