Environmental Fate · 3 件

TERRESTRIAL FATE: Based on a classification scheme(1), an estimated Koc value of 115(SRC), determined from a structure estimation method(2), indicates that 4-methylpyridine is expected to have high mobility in soil(SRC). The pKa of 4-methylpyridine is 5.98(3), indicating that this compound will exist partially in cation form in the environment and cations generally adsorb more strongly to soils containing organic carbon and clay than their neutral counterparts(4). Soil studies with 2-methylpyridine(5), a compound expected to have similar sorption properties as 4-methylpryidine(SRC), demonstrated that Koc can vary with pH with lowest adsorption occurring in the non-ionized form(5). Volatilization of 4-methylpyridine from moist soil surfaces is expected to be an important fate process(SRC) given a Henry's Law constant of 6.0X10-6 atm-cu m/mole(6). 4-Methylpyridine is expected to volatilize from dry soil surfaces(SRC) based upon a vapor pressure of 5.77 mm Hg at 25 °C(7). 4-Methylpyridine may biodegrade fairly rapidly in aerobic soil; however, under anaerobic conditions, this compound is expected to be persistent(8,9). 71.7 and 100% of the available nitrogen was released as inorganic nitrogen within 16 and 32 days following the incubation of 4-methylpyridine in aerobic Fincastle silt loam(7). 4-Methylpyridine added to an unpolluted surface soil under denitrifying or sulfidogenic conditions was 40 to 50% degraded in 3 months(8). Acclimation of microbes may be important as demonstrated by complete degradation of 4-methylpyridine in 2 weeks in polluted soil versus 3 months in unpolluted soil(9).

AQUATIC FATE: Based on a classification scheme(1), an estimated Koc value of 115(SRC), determined from a structure estimation method(2), indicates that 3-methylpyridine is not expected to adsorb to suspended solids and sediment(SRC). Volatilization from water surfaces is expected(3) based upon a Henry's Law constant of 6.0X10-6 atm-cu m/mole(4). Using this Henry's Law constant and an estimation method(3), volatilization half-lives for a model river and model lake are 6 and 47 days, respectively(SRC). According to a classification scheme(5), an estimated BCF of 3(SRC), from its log Kow of 1.22(6) and a regression-derived equation(2), suggests the potential for bioconcentration in aquatic organisms is low(SRC). 4-Methylpyridine may biodegrade fairly rapidly in aerobic waters; however, under anaerobic conditions, this compound is expected to be persistent. Complete biodegradation of 4-methylpyridine was reported in 2 to 4 days following incubation in acclimated natural waters(7). No biodegradation was reported over 200 days when 4-methylpyridine was incubated in sulfidogenic estuarine sediment(8). 4-Methylpyridine is not expected to undergo hydrolysis in the environment due to the lack of functional groups that hydrolyze under environmental conditions(3). The UV absorption spectrum of 4-methylpyridine in aqueous solution shows no absorption >290 nm(9); therefore, 4-methylpyridine is not expected to be susceptible to direct photolysis by sunlight(SRC).

ATMOSPHERIC FATE: According to a model of gas/particle partitioning of semivolatile organic compounds in the atmosphere(1), 4-methylpyridine, which has a vapor pressure of 5.77 mm Hg at 25 °C(2), is expected to exist solely as a vapor in the ambient atmosphere. Vapor-phase 4-methylpyridine is degraded in the atmosphere by reaction with photochemically-produced hydroxyl radicals(SRC); the half-life for this reaction in air is estimated to be 6 days(SRC), calculated from its rate constant of 2.69X10-12 cu cm/molecule-sec at 25 °C(3). The UV absorption spectrum of 4-methylpyridine in aqueous solution shows no absorption >290 nm(4); therefore, 4-methylpyridine is not expected to be susceptible to direct photolysis by sunlight(SRC).

Food Survey Values4-Methylpyridine was identified as a volatile component of fried bacon(1). 4-Methylpyridine is reported in pork meat volatiles(2).
Adverse EffectsNeurotoxin - Other CNS neurotoxin
Exposure RoutesThe substance can be absorbed into the body by inhalation, through the skin and by ingestion.

Metabolism/Metabolites · 2 件

Although the exact mechanism has not been determined, it appears that all methylpyridines are oxidized to their respective aromatic acids. ...Formation of an N-oxide also occurs with 4-methylpyridine.

The biotransformations of gamma-picoline were investigated in the rat. Wistar rats were admin gamma-picoline by stomach tube at a dose of 300 mg/kg body weight. Urine was collected for 24 hours in the presence of acetic acid while expired air was sampled in 5% aqueous sulfuric acid. gamma-Picoline and metabolites were extracted from the samples and characterized by chromatographic analysis and mass spectrometry. Unmetabolized gamma-picoline was identified in both expired air and 24 hour urine; excreted amounts were approx 2.5% in each. The mass spectral characteristics of gamma-picoline-N-oxide, methylisonicotinate and methylisonicotinurate in the urine were examined; excreted amounts were 1.5%, 50% and 5% respectively. The /results suggest/ that two metabolic pathways for gamma-picoline are observed: N-oxidation leads to the formation of N-oxide; and methyloxidation leads to the formation of isonicotinic acid excreted mainly unchanged and partly as the glycine conjugate isonicotinuric acid.

Absorption, Distribution and ExcretionIt was found that 4-picoline has a shorter residence time than the other substituted picolines in the liver, brain and kidney. Elimination occurred vua a biphasic process.

Pyridine, 4-methyl-: ACTIVE

Pyridine bases are a constituent of tars. They were isolated from coal tar or coal gas before synthetic manufacturing processes became established. The amounts contained in coal tar and coal gas are small, and the pyridine bases isolated from them are a mixture of many components. Thus, with a few exceptions, isolation of pure pyridine bases was expensive. Today, almost all pyridine bases are produced by synthesis. /Pyridine bases/

天然物の起源4-Methylpyridine is released by energy-related processes. It is present in coal gassification wastewater (Pellizzari et al 1979), the environment of coke ovens (Naizer and Mashek 1974) and in waters from shale oil waste production (Dobson et al 1985; Hawthorne et al 1985; Leenheer et al 1982). It is also present in coal tar (HSDB, 1988), cigarette smoke (Brunneman et al 1978; IARC 1976) and pyroligneous liquids from woods (Yasuhara and Sugiwara 1987). Methods for the biological treatment of wastewaters containing 4-methylpyridine have been developed (Roubickova 1986) and its movement through (Leenheer and Stuber 1981) and degradation (Sims and Somners 1985) in soils examined.

使用4-Methylpyridine is used to manufacture isonicotinic acid and derivatives, in waterproofing agents for fabric, and as a solvent for resins, pharmaceuticals, dyestuffs, rubber accelerators, and pesticides. It is also used as a catalyst and curing agent.

一般的な説明4-methyl pyridine is a colorless moderately volatile liquid. (NTP, 1992)

調製方法 · 2 件

Currently, 4-methylpyridine is produced by vapor-phase condensation of acetaldehyde and ammonia (3:1) with subsequent isolation of 4-methylpyridine from the reaction mixture. Reactants are exposed to dehydration-dehydrogenation catalysis such as lead oxide, copper oxide on alumina, thorium oxide, zinc oxide or cadmium oxide on silica-alumina, or cadmium fluoride on silica-magnesia at 400-500°C. This results in a 60% yield of 4-methylpyridine which is isolated by fractional distillation (USEPA 1982). Another production method involves the isolation from by-products of coking operations. The crude pyridine extracts come from noncondensable and condensable coke-oven gasses that have been dehydrated and separated by fractional distillation, but only 45% of 4-methylpyridine is obtained (USEPA 1982). 4-Methylpyridine also can be isolated from a dry distillation of bones or coal (Hawley 1977).

純化方法It can be purified as for 2-methylpyridine. Biddescombe and Handley's method (above) for 3-methylpyridine is also applicable. Lidstone [J Chem Soc 242 1940] purified it via the oxalate (m 137-138o) by heating 100mL of 4-methylpyridine to 80o and adding slowly110g of anhydrous oxalic acid, followed by 150mL of boiling EtOH. After cooling and filtering, the precipitate is washed with a little EtOH, then recrystallised from EtOH, dissolved in the minimum quantity of water and distilled with excess 50% KOH. The distillate is dried with solid KOH and again distilled. Hydrocarbons can be removed from 4-methylpyridine by converting the latter to its hydrochloride, crystallising from EtOH/diethyl ether, regenerating the free base by adding alkali and distilling. As a final purification step, 4-methylpyridine can be fractionally crystallised by partial freezing to effect a separation from 3-methylpyridine. Contamination with 2,6-lutidine is detected by its strong absorption at 270nm. The hydrochloride has m 161o, and the picrate has m 167o(from Me2CO, EtOH or H2O).
