化学的特性

Morpholine is a colorless to yellow liquid with a weak ammonia or fish-like odor. The odor threshold is 0.01 ppm. The reactivity of morpholine is mainly due to its secondary amine group. It readily undergoes organic condensations, alkylations, and arylations, resulting in the formation of various N-substituted morpholine compounds. Ethers are relatively chemically inert, hence the oxygen is of relatively little consequence except as a member of the heterocyclic ring (Texaco Chemical Co. 1982).

応急処置EYES: First check the victim for contact lenses and remove if present. Flush victim's eyes with water or normal saline solution for 20 to 30 minutes while simultaneously calling a hospital or poison control center. Do not put any ointments, oils, or medication in the victim's eyes without specific instructions from a physician. IMMEDIATELY transport the victim after flushing eyes to a hospital even if no symptoms (such as redness or irritation) develop.SKIN: IMMEDIATELY flood affected skin with water while removing and isolating all contaminated clothing. Gently wash all affected skin areas thoroughly with soap and water. IMMEDIATELY call a hospital or poison control center even if no symptoms (such as redness or irritation) develop. IMMEDIATELY transport the victim to a hospital for treatment after washing the affected areas.INHALATION: IMMEDIATELY leave the contaminated areatake deep breaths of fresh air. If symptoms (such as wheezing, coughing, shortness of breath, or burning in the mouth, throat, or chest) develop, call a physician and be prepared to transport the victim to a hospital. Provide proper respiratory protection to rescuers entering an unknown atmosphere. Whenever possible, Self-Contained Breathing Apparatus (SCBA) should be usedif not available, use a level of protection greater than or equal to that advised under Protective Clothing.INGESTION: DO NOT INDUCE VOMITING. Corrosive chemicals will destroy the membranes of the mouth, throat, and esophagus and, in addition, have a high risk of being aspirated into the victim's lungs during vomiting which increases the medical problems. If the victim is conscious and not convulsing, give 1 or 2 glasses of water to dilute the chemical and IMMEDIATELY call a hospital or poison control center. IMMEDIATELY transport the victim to a hospital. If the victim is convulsing or unconscious, do not give anything by mouth, ensure that the victim's airway is open and lay the victim on his/her side with the head lower than the body. DO NOT INDUCE VOMITING. Transport the victim IMMEDIATELY to a hospital. (NTP, 1992)General First Aid:· Call 911 or emergency medical service.· Ensure that medical personnel are aware of the material(s) involved, take precautions to protect themselves and avoid contamination.· Move victim to fresh air if it can be done safely.· Administer oxygen if breathing is difficult.· If victim is not breathing:-- DO NOT perform mouth-to-mouth resuscitationthe victim may have ingested or inhaled the substance.-- If equipped and pulse detected, wash face and mouth, then give artificial respiration using a proper respiratory medical device (bag-valve mask, pocket mask equipped with a one-way valve or other device).-- If no pulse detected or no respiratory medical device available, provide continuous compressions. Conduct a pulse check every two minutes or monitor for any signs of spontaneous respirations.· Remove and isolate contaminated clothing and shoes.· For minor skin contact, avoid spreading material on unaffected skin.· In case of contact with substance, remove immediately by flushing skin or eyes with running water for at least 20 minutes.· For severe burns, immediate medical attention is required.· Effects of exposure (inhalation, ingestion, or skin contact) to substance may be delayed.· Keep victim calm and warm.· Keep victim under observation.· For further assistance, contact your local Poison Control Center.· Note: Basic Life Support (BLS) and Advanced Life Support (ALS) should be done by trained professionals.Specific First Aid:· For corrosives, in case of contact, immediately flush skin or eyes with running water for at least 30 minutes. Additional flushing may be required.(General first aid procedures)Eye: Irrigate immediately - If this chemical contacts the eyes, immediately wash (irrigate) the eyes with large amounts of water, occasionally lifting the lower and upper lids. Get medical attention immediately.Skin: Water flush immediately - If this chemical contacts the skin, immediately flush the contaminated skin with water. If this chemical penetrates the clothing, immediately remove the clothing and flush the skin with water. Get medical attention promptly.Breathing: Respiratory supportSwallow: Medical attention immediately - If this chemical has been swallowed, get medical attention immediately.
安全保管Fireproof. Separated from strong oxidants and acids. Dry.
Firefighting HazardsVapor is heavier than air and may travel some distance to source of ignition and flash back.
暴露管理・個人防護· Wear positive pressure self-contained breathing apparatus (SCBA).· Wear chemical protective clothing that is specifically recommended by the manufacturer when there is NO RISK OF FIRE.· Structural firefighters' protective clothing provides thermal protection but only limited chemical protection.5.0 [ppm]
消火手順Use dry chemical, carbon dioxide, or alcohol foam extinguishers. Vapors are heavier than air and will collect in low areas. Vapors may travel long distances to ignition sources and flashback. Vapors in confined areas may explode when exposed to fire. Containers may explode in fire. Storage containers and parts of containers may rocket great distances, in many directions. If material or contaminated runoff enters waterways, notify downstream users of potentially contaminated waters. Notify local health and fire officials and pollution control agencies. From a secure, explosion-proof location, use water spray to cool exposed containers. If cooling streams are ineffective (venting sound increases in volume and pitch, tank discolors or shows any signs of deforming), withdraw immediately to a secure position ... The only respirators recommended for fire fighting are self-contained breathing apparatuses that have full facepieces and are operated in a pressure-demand or other positive-pressure mode.If material on fire or involved in fire: Do not extinguish fire unless flow can be stopped. Use water in flooding quantities as fog. Solid streams of water may be ineffective. Cool all affected containers with flooding quantities of water. Apply water from as far a distance as possible. Use "alcohol" foam, carbon dioxide or dry chemical.Use flooding quantities of water as fog or spray, dry chemical, "alcohol resistant" foam, or carbon dioxide. Use water spray to keep fire-exposed containers cool.
保管条件Before entering confined space where this chemical may be present, check to make sure that an explosive concentration does not exist. Morpholine must be stored to avoid contact with strong acids, such as nitric acid, and strong oxidizers, such as chlorine dioxide, bromine, nitrates, and permanganates, since violent reactions occur.Separate from oxidizing materials and acids. Store in a cool, dry, well ventilated location. Outside or detached storage is preferred. Inside storage should be in a standard flammable liquids storage warehouse, room, or cabinet.Morpholine should be protected from atmospheric moisture and carbon dioxide. As morpholine corrodes copper, aluminum, zinc, and galvanized surfaces, the compound should be stored in iron or steel containers, preferably located outdoors, above ground, and surrounded by dykes to contain spills or leaks.Keep containers closed and store in a cool, dark place.Unstable if stored in copper or zinc containers.
清掃方法Spill handling: evacuate and restrict persons not wearing protective equipment from area of spill or leak until cleanup is complete. Remove all ignition sources. Establish forced ventilation to keep levels below explosive limit. Absorb liquids in vermiculite, dry sand, earth, peat, carbon, or a similar material and deposit in sealed containers. Keep this chemical out of a confined space, such as a sewer, because of the possibility of an explosion, unless the sewer is designed to prevent the build-up of explosive concentrations. It may be necessary to contain and dispose of this chemical as a hazardous waste. If material or contaminated runoff enters waterways, notify downstream users of potentially contaminated waters. Contact your Department of Environmental Protection or your regional office of the federal EPA for specific recommendations.Environmental considerations - land spill: Dig a pit, pond, lagoon, holding area to contain liquid or solid material. /SRP: If time permits, pits, ponds, lagoons, soak holes, or holding areas should be sealed with an impermeable flexible membrane liner./ Dike surface flow using soil, sand bags, foamed polyurethane, or foamed concrete. Absorb bulk liquid with fly ash, cement powder, or commercial sorbents.Environmental considerations - water spill: Use natural barriers or oil spill control booms to limit spill travel. Remove trapped material with suction hoses.Environmental considerations - air spill: Apply water spray or mist to knock down vapors. For more Cleanup Methods (Complete) data for Morpholine (7 total), please visit the HSDB record page.

処分方法SRP: The most favorable course of action is to use an alternative chemical product with less inherent propensity for occupational harm/injury/toxicity or environmental contamination. Recycle any unused portion of the material for its approved use or return it to the manufacturer or supplier. Ultimate disposal of the chemical must consider: the material's impact on air qualitypotential migration in soil or watereffects on animal and plant lifeand conformance with environmental and public health regulations.Package lots. Distill for reuse or package and label for disposal by burning.Small quantities. Wear eye protection, laboratory coat, and rubber gloves. Dissolve the morpholine (1 mL) in 50 mL of 3 M sulfuric acid (prepared by slowly adding 8 mL of concentrated sulfuric acid to 21 mL or water). Weigh 10 g of potassium permanganate and stir small portions of the solid into the morpholine solution over a period of about 1 hr. Stir the mixture at room temperature for 48 hr, and then neutralize the solution by adding solid sodium carbonate or a 10% solution of sodium hydroxide. Add solid sodium bisulfite until solution is colorless. Decant the clear liquid into the drain and discard any brown solid with regular refuse.Incineration is acceptable and the preferred method of disposalhowever, nitrogen oxide emission controls may be required to meet environmental regulations. Morpholine is also broken down by activated sludge and this is a possible method of disposal under controlled conditions. For more Disposal Methods (Complete) data for Morpholine (7 total), please visit the HSDB record page.
相互作用A group of seven female Sprague-Dawley rats /were fed/ 5 g morpholine together with 5 g nitrite/kg diet for 12 weeks. After 39 weeks, all of the animals developed hepato-cellular adenomas, six out of the seven hepatocellular carcinomas, two hemangioendotheliomas of the liver, one a cyst-adenocarcinoma of the liver, and one a renal adenoma. Rats fed morpholine or nitrite alone did not develop tumors.Pregnant rats /were fed/ on a diet containing various concentrations of morpholine, sodium nitrite, and NMOR (N-nitrosomorpholine) ... Hepatocellular carcinoma and hemangio-sarcomas of the liver and angiosarcoma of the lungs were the most common tumors observed in the rats. The neoplasms induced by nitrite and morpholine were morphologically similar to those induced by NMOR. High concentrations (1000 mg/kg) of morpholine and nitrite together were carcinogenic to rats. When the morpholine concentration was reduced and the nitrite concentration remained high, the incidence of hepatic cell carcinomas decreased with a linear dose-response relationship. When morpholine concentration remained high, with decreasing nitrite concentration, the number of hepatic tumors was sharply reduced. This agrees with the observation in vitro that the nitrosation of morpholine depends on the square of the nitrite concentration.Groups of 40 male MRC Wistar rats were treated for 2 yr with either 10 g morpholine/kg diet and drinking-water containing 3 g sodium nitrite/L, or with drinking-water containing 0.15 g NMOR/L. In both cases, one group of rats was also given sodium ascorbate (22.7 g/kg diet). The results of treatment with morpholine plus nitrite or with NMOR were similar to those reported /in the feeding experiments/. When ascorbate was present, the liver tumors induced by morpholine plus nitrite had a longer induction period (93 vs 54 weeks) and a slightly lower incidence (49% vs 65%). However, ascorbate did not affect liver tumor induction by preformed NMOR. Of those treated with morpholine, nitrite, and ascorbate, 21/39 animals developed forestomach tumors. In a similar study using Syrian hamsters, a high morpholine, high nitrite diet (1000 mg/kg) induced liver cancer in some animals. In contrast, hamsters fed NMOR in the diet seemed to have greater resistance to tumor induction.Bis(2,2-dimethyl-4-methanesulfonic acid sodium salt-1,2-dihydroquinoline)-6,6'-methane (MIDQ-DA) inhibited liver necrosis induced in rats by simultaneous admin of morpholine and sodium nitrite. For more Interactions (Complete) data for Morpholine (17 total), please visit the HSDB record page.

環境中動態TERRESTRIAL FATE: Based on a classification scheme(1), an estimated Koc value of 7.4(SRC), determined from a log Kow of -0.86(2) and a regression-derived equation(3), indicates that morpholine is expected to have very high mobility in soil(SRC). The pKa of morpholine is 8.49(4), indicating that this compound will partially exist in the cation form in the environment and cations generally adsorb more strongly to soils containing organic carbon and clay than their neutral counterparts(5). Volatilization of cationic morpholine from moist soil surfaces will not occur since ions do not volatilize(SRC). Volatilization of the neutral form of morpholine from moist soil surfaces may be an important fate process(SRC) given an estimated Henry's Law constant of 1.2X10-6 atm-cu m/mole(SRC), derived from its vapor pressure, 10.1 mm Hg(6), and assigned value for water solubility of 1.0X10+6 mg/L (miscible)(6). Morpholine is expected to volatilize from dry soil surfaces(SRC) based upon its vapor pressure(6). The theoretical BOD using activated sludge in the Japanese MITI test was 0% after 2 weeks(7). Results from longer biodegradation screening tests indicate morpholine is biodegradable in activated sludge with degradation rates of 87% to greater than 90% within 28 to 31 days, respectivelylag periods ranging from 7 to 21 days were observed for these tests(8,9). In a study investigating morpholine-degrading bacteria, it was noted that these bacteria were slow to become established, and could only be done so in biological treatment plants with a mean solids retention times of more than 8 days(10). Therefore, morpholine is expected to be biodegradable in environments where morpholine-degrading organisms are present and after acclimation to these organism is achieved(SRC).AQUATIC FATE: Based on a classification scheme(1), an estimated Koc value of 7.4(SRC), determined from a log Kow of -0.86(2) and a regression-derived equation(3), indicates that morpholine is not expected to adsorb to suspended solids and sediment(SRC). A pKa of 8.49(4) indicates morpholine will exist almost entirely in the cation form at pH values of 5 to 9 and therefore volatilization from water surfaces is not expected to be an important fate process(5). According to a classification scheme(6), measured BCF values of less than 0.3 to 0.65 and less than 2.8(7), suggest that bioconcentration in aquatic organisms is low(SRC). Only 3 out of 58 river die-away tests failed to biodegrade morpholine or failed to contain morpholine-degrading bacteria. Lag periods of approximately 6 to 50 days were observed(8). These data indicate that biodegradation in aquatic environments will vary depending on conditions(SRC).ATMOSPHERIC FATE: According to a model of gas/particle partitioning of semivolatile organic compounds in the atmosphere(1), morpholine, which has a vapor pressure of 10.1 mm Hg at 25 °C(2), is expected to exist solely as a vapor in the ambient atmosphere. Vapor-phase morpholine is degraded in the atmosphere by reaction with photochemically-produced hydroxyl radicals(SRC)the half-life for this reaction in air is estimated to be 2.8 hours(SRC), calculated from its rate constant of 1.4X10-10 cu cm/molecule-sec at 25 °C(SRC) that was derived using a structure estimation method(3). Morpholine does not contain chromophores that absorb at wavelengths >290 nm(4) and therefore is not expected to be susceptible to direct photolysis by sunlight(SRC).
食品調査値An average morpholine concentration in baked ham obtained from a local market (n = 5) in Kobe City, Japan was 0.20 ppm, testing date not specified. It was not detected in fish sausage (n = 5), cod roe (n = 3), spinach (n = 2 ), or miso (n = 2) (detection limit = 0.01 ppm)(1). During a study analyzing naturally-occurring secondary amines in locally purchased food, morpholine was detected in the following food items at concentrations of ppm: 0.5 in baked ham, 0.4 in frankfurters, 0.2 in evaporated milk, 1 in coffee (average of two brandsper dry weight), trace amounts (<0.1) in tea, 0.4 in canned beer, <0.2 in bottled beer, and <0.7 in wine(2).

Algal growth /in the same species was evaluated/ by measuring in vivo the fluorometric units at 48, 72, 96 hr, and 7 days. A 96 hr EC50 of 28 mg/L was determined./AQUATIC SPECIES/ Exposure of rainbow trout to 5.7 times 10 (-10) M (5 times 10 (-5) mg/L) morpholine during smolting period did not influence the subsequent neural response to this substance when tested immediately and 12 months after smolting. Perfusion of the olfactory organs with 1% morpholine for 10 min irreversibly inhibited the olfactory responsiveness to odorants./PLANTS/ 4.4 +/- 0.9 mol morpholine/cu m (383 mg/L) lowers the percentage germination of lettuce seeds (Lactuca sativa) by 50% of the control value. The seeds were kept on agar for 3 days in sealed plastic containers at a temperature of 30 °C.
Plant ConcentrationsMorpholine was detected in the medicinal plant roots obtained from a local market in Ibadan, Nigeria(1).

Medical SurveillanceThe following medical procedures should be made available to each employee who is exposed to morpholine at potentially hazardous levels: 1. Initial Medical Screening: Employees should be screened for history of certain medical conditions which might place the employee at increased risk from morpholine exposure. /These include/ chronic respiratory disease ... liver disease ... kidney disease ... eye disease ... /and/ skin disease. 2. Periodic Medical Examination: Any employee developing the above-listed conditions should be referred for further medical examination.
大気基準This action promulgates standards of performance for equipment leaks of Volatile Organic Compounds (VOC) in the Synthetic Organic Chemical Manufacturing Industry (SOCMI). The intended effect of these standards is to require all newly constructed, modified, and reconstructed SOCMI process units to use the best demonstrated system of continuous emission reduction for equipment leaks of VOC, considering costs, non air quality health and environmental impact and energy requirements. Morpholine is produced, as an intermediate or a final product, by process units covered under this subpart.

使用

Morpholine is made by dehydrating ethanolamines. Its main use is as a rubber accelerator in manufacturing tires. This process requires high temperature (300°F) and pressure, which increase the hazards. Morpholine is also used as a boiler water additive, brightener for detergents, and corrosion inhibitor, in the preservation of book paper, in waxes and polishes, and in organic synthesis.

一般的な説明

An aqueous solution with a fishlike odor. Corrosive to tissue and moderately toxic by ingestion and inhalation.

製法・合成法

調製方法

Morpholine is produced by reacting diethylene glycol, ammonia, and a small amount of hydrogen over a hydrogenation catalyst at 150-400°C and 30-400 atmospheres with the morpholine being recovered by fractional distillation. Various byproducts include 2-(2-aminoethoxy)ethanol and Af-alkylmorpholines (NRC 1981).

純化方法

Dry morpholine with KOH, fractionally distil it, then reflux it with Na, and again fractionally distil it. Dermer & Dermer [J Am Chem Soc 59 1148 1937] precipitated it as the oxalate by adding slowly to slightly more than 1 molar equivalent of oxalic acid in EtOH. The precipitate is filtered off and recrystallised twice from 60% EtOH [1:1 salt has m 190-195o(dec)]. Addition of the oxalate to concentrated aqueous NaOH regenerated the base, which is separated and dried with solid KOH, then sodium, before being fractionally distilled. The hydrochloride has m 178-179o (from MeOH/Et2O), and the picrate has m 151.6o (from aqueous EtOH). [Beilstein 27 II 3, 27 III/IV 15.]
