Non-Human Toxicity Excerpts
/LABORATORY ANIMALS: Acute Exposure/ /Ten Sprague-Dawley rats, both male and female aged 5-8 weeks old (bw 136-153 g males, 136-146 g females) were given a single dose (2000 mg/kg bw) of 2,2-bis(bromomethyl)-1,3-propanediol in Arachis oil by oral gavage/ ... Two females died 30 min after dosing. One male died one hr after dosing ... Common signs of systemic toxicity noted were coma, labored respiration, hunched posture, and lethargy with additional signs of ataxia, ptosis, and decreased respiratory rate. Isolated incidents of loss of righting reflex were noted in two animals. Surviving animals appeared normal one or two days after dosing. The acute median lethal dose (LD50) was found to be greater than 2000 mg/kg bw.
/LABORATORY ANIMALS: Acute Exposure/ Male and female CD-1 rats (35 days old) were given 2,2-bis(bromomethyl)-1,3-propanediol by oral gavage at doses of 1247, 1484, 1765, 2101, 2500 mg/kg/ ... Initial signs of reaction to treatment consisted of a rapid progression from reduced motor activity, ataxia, prone posture and prostration to a loss of consciousness between 15 and 30 min after administration. Coma and death, between one and six hr after dosing, followed loss of consciousness in a total of 22 animals. The remaining animals generally regained consciousness one to two hours after administration of the lowest dosage, but recovery was less rapid as the dosage increased. All surviving animals were asymptomatic from Day 3 and all showed normal bodyweight gains over the 14-day observation period. From the observed mortality data, the acute median lethal dosage (LD50) and 95% CI of Dinol, calculated by probit analysis were found to be 1880 (1691-2120) mg/kg.
/LABORATORY ANIMALS: Acute Exposure/ Upon single dermal application /of 2,2-bis(bromomethyl)propane-1,3-diol 0.5 g on New Zealand White rabbits/, a barely discernible erythematous response on the abraded skin of one animal at 72 hr account for a Primary Irritation Index of zero. Bandage burn was recognized on 3 animals and slight exfoliation was present in one animal on day 8. A single dermal application of test substance produced a Primary Irritation Index of zero, thus placing the material into the lowest available class of 'mildly irritating' to the skin.
/LABORATORY ANIMALS: Acute Exposure/ A single installation of 100 mg of 2,2-bis(bromomethyl)propane-1,3-diol into the conjunctival sac of six New Zealand White rabbits provoked corneal and conjunctival lesions in four animals regarded as positive reactions under this method. Therefore the test substance was considered to elicit a positive result as an eye irritant. The positive reaction had resolved by Day 8 and may therefore be considered as reversible. / One animal displayed diffuse crimson redness of the conjunctivae in addition to the corneal lesions. All animals exhibited slight conjunctivitis at the 24, 48, and 72-hr readings. This persisted to Day 8 in two animals. Immediate pain responses upon instillation of the test material ranged from practically no initial pain to moderate initial pain/.

Metabolism/Metabolites
In a 2-year bioassay 2,2-bis(bromomethyl)-1,3-propanediol (BMP) was shown to be a multisite carcinogen in rats and mice. Because glucuronidation is the key metabolic transformation of BMP by rats, in this study the in vitro hepatic glucuronidation of BMP was compared across several species. In addition, the glucuronidation activities of human intestinal microsomes and specific human hepatic UDP-glucuronosyltransferase (UGT) enzymes for BMP were determined. To explore other possible routes of metabolism for BMP, studies were conducted with rat and human hepatocytes. Incubation of hepatic microsomes with BMP in the presence of UDP-glucuronic acid resulted in the formation of a BMP monoglucuronide. The order of hepatic microsomal glucuronidation activity of BMP was rats, mice >> hamsters > monkeys >>> humans. The rate of glucuronidation by rat hepatic microsomes was 90-fold greater than that of human hepatic microsomes. Human intestinal microsomes converted BMP to BMP glucuronide at a rate even lower than that of human hepatic microsomes. Among the human UGT enzymes tested, only UGT2B7 had detectable glucuronidation activity for BMP. BMP monoglucuronide was the only metabolite formed when BMP was incubated with suspensions of freshly isolated hepatocytes from male F-344 rats or with cryopreserved human hepatocytes. Glucuronidation of BMP in human hepatocytes was extremely low. Overall, the results support in vivo studies in rats in which BMP glucuronide was the only metabolite found. The poor glucuronidation capacity of humans for BMP suggests that the pharmacokinetic profile of BMP in humans will be dramatically different from that of rodents.
Pentaerythritol Dibromide has known human metabolites that include (2R,3R,4R,5S,6S)-6-[2,2-Bis(bromomethyl)-3-hydroxypropoxy]-3,4,5-trihydroxyoxane-2-carboxylic acid.
Absorption, Distribution and Excretion
... Studies were performed to characterize the dispositional and metabolic fate of 2,2-bis(bromomethyl)-1,3-propanediol (BMP) after oral or intravenous administration to male Fischer-344 rats. After a single oral administration of (14)C-BMP (10 or 100 mg/kg) >80% of the low dose and 48% of the high dose were excreted by 12 hr in the urine predominantly as a glucuronide metabolite. After repeated daily oral doses for 5 or 10 days, route and rate of elimination were similar to those obtained after single administrations of BMP. In all studies, the radioactivity recovered in feces was low (<15%). The total amount of radioactivity remaining in tissues at 72 hr after a single oral administration of BMP (100 mg/kg) was less than 1% of the dose, and repeated daily dosing did not lead to retention in tissues. After intravenous administration, the radiolabel found in blood decreased rapidly. Excretion profiles were similar to those after oral administration. Parent BMP and BMP glucuronide were present in blood plasma after oral or intravenous dosing. After an intravenous dose of BMP (15 mg/kg) the hepatic BMP glucuronide was primarily exported into the bile (>50% within 6 hr), but it underwent enterohepatic recycling with subsequent elimination in the urine. These data indicate that the extensive extraction and rapid glucuronidation by the liver limits exposure of internal tissues to BMP by greatly reducing its systemic bioavailability after oral exposure.
Analysis of selected tissues indicated an increase in bromide content in tissues of rats ingesting 100 mg FR 1138/kg/day. At 5 mg FR 1138/kg/day, there was only a marginal increase in bromide content of some tissues, with most values in same range as controls.

1,3-Propanediol, 2,2-bis(bromomethyl)-: ACTIVE
The mechanism and kinetics of the spontaneous decomposition of 2,2-bis(bromomethyl)propan-1,3-diol (DBNPG) and its decomposition daughter products were determined in aqueous solution at a temperature range between 30 and 70 °C and pH from 7.0 to 9.5. DBNPG decomposition in basic aqueous solutions involves release of bromide ions through a sequential formation of 3-bromomethyl-3-hydroxymethyloxetane (BMHMO) and 2,6-dioxaspiro[3.3]heptane (DOH). DBNPG decomposition into BMHMO is a two-stage reaction. The first stage is an acid/base equilibrium, in which an alkoxide is formed. In the second stage, DBNPG predominantly undergoes an intramolecular nucleophilic substitution to form the BMHMO. The transformation rate increases with the pH and the energy barrier for the degradation is 98 kJ/mol. Good agreement was found between the rate coefficients derived from variations in the organic molecules concentrations and those determined from the changes in the Br(-) concentration ...
