Mechanism of Action · 3건

Hepatic enzyme induction (T4 conjugation) leads to increased excretion of T4, compensatory activation of the pituitary, increased serum TSH concentration and activation of the thyroid. Depending on the magnitude of the T4-decrease, the feed-back system may manage to produce sufficient amounts of T3, or if the reduction is severe, lead to a condition of hypothyroidism. This MoA is based on the assumption that hepatic enzymes involved in the metabolism of T4/T3 are induced by HBCDD ... /One study/ has indeed identified induction of T4-UGT transferase by HBCDD, and the induction occurred in both sexes. The hypothesis postulates that the first effect (occurring at the lowest exposure level) is enzyme induction, followed by activation of the pituitary (resulting in TSH synthesis), activation of the thyroid (hyperactive cells/weight increase), and if the thyroid is incapable of producing sufficient amounts of T4/T3, effects in other tissues/systems regulated by T4/T3. In females, the BMD-L for induction of T4-UGT is 4.1 mg/kg/day (and LBD/CYP3A4 is induced as from 10 mg/kg/day), the BMD-L for pituitary weight 29.9 mg/kg/day, the (uncertain) BMD-L for thyroid weight 1.6 mg/kg/day (the weight increase seems undisputable at 30 mg/kg/day), and there are histological signs of thyroid hyperactivity at 25, 47, and 177 mg/kg/day for nuclear size, cell height, and vacuolization, respectively. Considering the variation in all data, this sequence of events could support the theory with regard to females. In males, enzyme induction occurs (the uncertain BMD-L for T4-UGT transferase is 0.1 mg/kg/day; PROD/CYP2B is induced as from 10 mg/kg/day), there are no effects of T4 or the pituitary, but there are histological signs of thyroid hyperactivity at 39, 90, and 199 mg/kg/day for nuclear size, vacuolization, and follicle size, respectively. There are no effects on the thyroid weight.

Considering that in vitro data indicates an interaction of HBCDD with many different hormone systems, it is possible that HBCDD also could affect the thyroid system via other mechanisms than hepatic enzyme induction. In cell cultures, HBCDD was found to exert antagonistic effects at the progesterone receptor, androgen receptor, and estrogen receptor (IC50 1-5 uM for gamma-HBCDD). A low binding of alpha-HBCDD to the thyroxin-binding transport protein TTR was also indicated in vitro (IC50 12 uM). Alpha-HBCDD was a T3-agonist (21 % of maximal T3-effect at 1 uM alpha-HBCDD) and enhanced T3-dependent effects both in rat cells and ex vivo in tadpole tail tips. It is not likely that these in vitro effects can explain all in vivo effects on the thyroid system, but theoretically they could be factors contributing to effects on the thyroid hormone homeostasis. For instance, the binding of HBCDD (or metabolites of HBCDD) to TTR could displace T4 from TTR, making T4 more susceptible to metabolism and excretion. Binding of chemicals to TTR is usually increased by hydroxy-groups (the endogenous ligand thyroxin is hydroxylated), and one may speculate that hydroxy-HBCDD could have a higher binding affinity to TTR than HBCDD itself. OH-HBCDD is formed from HBCDD by microsomes in vitro, but it is not known if here is any sex-dependent difference in formation of OH-HBCDD. A T3-potentiating effect could possibly occur simultaneously as the concentration of T4 is decreased, masking some of the hypothyroidogenic symptoms.

Median hexabromocyclododecane plasma levels in 2002 from 10 pregnant women living in Bodo, Norway were 19, 7, and 23 pg/mL plasma for the alpha-, beta-, and gamma-isomers, respectively; levels of plasma samples from 10 women living in Taimyr, Russia were 21, 8, and 33 pg/mL plasma, respectively(1). Hexabromocyclododecane levels in mothers' milk from the Stockholm, Sweden area increased from 0.13 pmol/g lipid in 1980 to 0.60 pmol/g lipid in 2004, with the highest levels reported in 2001 and 2002, 0.83 and 0.93 pmol/g lipid, respectively. Levels in Norwegian breast milk samples ranged from 0.25-2 ng/g lipids over a study period of 1986-2001(1).

The National Food Administration in Sweden has analyzed HBCDD and other substances in mother's milk from Swedish women in two different studies. The first study detected HBCDD in 12 of 33 samples. The samples were taken from primiparous, age 19-40, living in Uppsala County, Sweden, two weeks after delivery. The limit of detection was /leas than/ 15 pg HBCDD/g mothers milk (fresh weight). On a lipid weight basis, the mean and max concentrations were 0.45 and 2.4 ng/g fat, respectively(1).

Hexabromocyclododecane (HBCD) is a type of brominated flame retardant. We investigated the time trend of the stereoisomer-specific concentrations of HBCD (alpha, beta, and gamma) in breast milk samples of Japanese women by using liquid chromatography-tandem mass spectrometry (LC/MS/MS). The pooled milk lipid samples collected in 1973, 1978, 1983, 1988, 1993, 1999, and from 2000 to 2006 were analysed. The concentration was below the detection limit in the samples collected between 1973 and 1983 and increased in those collected since 1988. Between 1988 and 2006, alpha-HBCD was detected in all 11 breast milk samples collected from 25 to 29-year-old Japanese women; gamma-HBCD, in 7; and beta-HBCD, in none of the samples. Time trend of summation operatorHBCD appeared to be related to that of the industrial HBCD consumption level in Japan.

Concentrations of tetrabromobisphenol A (TBBPA) and alpha-, beta-, and gamma-isomers of hexabromocyclododecanes (HBCDs) were determined by liquid chromatography-tandem mass spectrometry (LC-ESI-MS/MS) in human adipose tissue obtained in New York City. ... The overall mean concentrations (mean+/-SD) of TBBPA and HBCDs were 0.048+/-0.102 and 0.333+/-0.571 ng/g lipid wt in human adipose tissue samples. ... Overall mean concentrations of HBCDs were 5-10-fold higher than mean TBBPA concentrations, in all of the samples analyzed. ... In the human adipose tissue samples, the concentrations of HBCDs were 3-4 orders of magnitude lower than the concentrations of polybrominated diphenyl ethers (PBDEs) previously reported for the same set of tissue samples. Concentrations of HBCDs in human samples from the United States were 1-5-fold lower than the concentrations reported from several European countries.

Thyroid hormone (TH) plays an essential role in growth and differentiation of the central nervous system. Deficiency of TH during perinatal period results in abnormal brain development known as cretinism in human. We recently reported that an environmental chemical 1,2,5,6,9,10-a-hexabromocyclododecane (HBCD) suppressed TH receptor (TR)-mediated transcription. To examine the effect of HBCD on cerebellar granule cells, we used purified rat cerebellar granule cells in reaggregate culture. Low dose HBCD (10(-10)M) significantly suppressed TH-induced neurite extension of granule cell aggregate. To clarify further the mechanisms of such suppression, we added brain-derived neurotrophic factor (BDNF) into culture medium, since BDNF plays a critical role in promoting granule cell development and is regulated by TH. BDNF completely rescued HBCD-induced suppression of granule cell neurite extension in the presence of T3. These results indicate that HBCD may disrupt TH-mediated brain development at least in part due to a disruption of the T3 stimulated increase in BDNF and BDNF may possess ability to ameliorate the effect of HBCD in granule cells.

Hexabromocyclododecane (HBCD), a type of brominated flame retardants (BFR), has become ubiquitous organic contaminants in recent years. However, studies on HBCD toxicity and the related molecular mechanisms are so far limited. The objective of the present study was to investigate the effects of oligomeric proanthocyanidins (OPCs) on cytotoxicity induced by HBCD and the underlying molecular mechanisms. HepG2 cells were treated with HBCD and/or OPCs at different concentrations, and cell viability, cell apoptosis, reactive oxygen species (ROS) production, cellular Ca(2+) level, mitochondrial membrane potential, cytochrome C (Cyt-c) release, and nuclear factor-erythroid 2-related factor 2 (Nrf2) proteins expression were evaluated. Results showed that HBCD induced toxic effects in HepG2 cells in a concentration-dependent manner. HBCD at high concentrations (40 and 60 uM) caused a significant decrease of cell viability and led to elevated cell apoptosis ratio, intracellular Ca(2+) level, cytoplasmic Cyt-c level, and ROS production, together with a loss of /mitochondrial membrane potential/ and mobilization of Nrf2. Pretreatment with OPCs effectively attenuated the cytotoxic effects and ROS production, as well as mitochondrial responses induced by HBCD. Thus, OPCs could alleviate cytotoxicity in HepG2 cells induced by HBCD through regulation on intracellular Ca(2+) level and ROS formation in a mitochondrial pathway.

Proteomic effect screening in zebrafish liver cells was performed to generate hypotheses regarding single and mixed exposure to the BFRs HBCD and TBBPA. Responses at sublethal exposure were analysed by two-dimensional gel electrophoresis followed by MALDI-TOF and FT-ICR protein identification. Mixing of HBCD and TBBPA at sublethal doses of individual substances seemed to increase toxicity. Proteomic analyses revealed distinct exposure-specific and overlapping responses suggesting novel mechanisms with regard to HBCD and TBBPA exposure. While distinct HBCD responses were related to decreased protein metabolism, TBBPA revealed effects related to protein folding and NADPH production. Overlapping responses suggest increased gluconeogenesis (GAPDH and aldolase) while distinct mixture effects suggest a pronounced NADPH production and changes in proteins related to cell cycle control (prohibitin and crk-like oncogene). We conclude that mixtures containing HBCD and TBBPA may result in unexpected effects highlighting proteomics as a sensitive tool for detecting and hypothesis generation of mixture effects.

Environmental Fate · 3건

TERRESTRIAL FATE: Based on a classification scheme(1), an estimated Koc value of 91,000(SRC), determined from a structure estimation method(2), indicates that hexabromocyclododecane is expected to be immobile in soil(SRC). Volatilization of hexabromocyclododecane from moist soil surfaces is expected to be an important fate process(SRC) given an estimated Henry's Law constant of 6.1X10-6 atm-cu m/mole(SRC), derived from its vapor pressure, 4.7X10-7 mm Hg(3), and water solubility, 0.0656 mg/L(3). However, adsorption to soil is expected to attenuate volatilization(SRC). Hexabromocyclododecane is not expected to volatilize from dry soil surfaces(SRC) based upon its vapor pressure(3). Results of screening studies(3) indicate that biodegradation of hexabromocyclododecane can range from not degradable to being inherently biodegradable depending on test conditions(SRC). Aerobic soil degradation tests have determined half-lives ranging from 63-441 days(3,4). Dissipation half-lives were roughly 2-4 times longer in biologically-inhibited soil indicating that biodegradation is an important fate process(3). One microcosm study found biotransformation half-lives of 63 and 6.9 days in aerobic and anaerobic soil, respectively(4).

AQUATIC FATE: Based on a classification scheme(1), an estimated Koc value of 91,000(SRC), determined from a structure estimation method(2), indicates that hexabromocyclododecane is expected to adsorb to suspended solids and sediment(SRC). Volatilization from water surfaces is expected(3) based upon an estimated Henry's Law constant of 6.1X10-6 atm-cu m/mole(SRC), derived from its vapor pressure, 4.7X10-7 mm Hg(4), and water solubility, 0.0656 mg/L(4). Using this Henry's Law constant and an estimation method(3), volatilization half-lives for a model river and model lake are 15.6 days and 122 days, respectively(SRC). However, volatilization from water surfaces is expected to be attenuated by adsorption to suspended solids and sediment in the water column. The estimated volatilization half-life from a model pond is >10 years if adsorption is considered(5). According to a classification scheme(6), measured BCF values of 18,200 in fathead minnows (Pimephales promelas)(7) and 8,974 in rainbow trout (Oncorhynchus mykiss)(8), suggest that bioconcentration in aquatic organisms is very high, provided the compound is not metabolized by the organism(SRC). Results of screening studies(4) indicate that biodegradation of hexabromocyclododecane can range from not degradable to being inherently biodegradable depending on test conditions(SRC). In a microcosm study using two river waters, the biotransformation half-lives of hexabromocyclododecane in aerobic and anaerobic river water were 11-32 and 1.1-1.5 days, respectively(9). Sediment half-lives of 101 and 191 days have been reported for 20 and 12 °C, respectively(10). Aqueous hydrolysis tests conducted over 39-day periods detected no hydrolysis of hexabromocyclododecane(4).

ATMOSPHERIC FATE: According to a model of gas/particle partitioning of semivolatile organic compounds in the atmosphere(1), hexabromocyclododecane, which has a vapor pressure of 4.7X10-7 mm Hg at 25 °C(2), will exist in both the vapor and particulate phases in the ambient atmosphere. Vapor-phase hexabromocyclododecane is degraded in the atmosphere by reaction with photochemically-produced hydroxyl radicals(SRC); the half-life for this reaction in air is estimated to be 2.6 days(SRC), calculated from its rate constant of 6.1X10-12 cu cm/molecule-sec at 25 °C(SRC) that was derived using a structure estimation method(3). Particulate-phase hexabromocyclododecane may be removed from the air by wet or dry deposition(SRC). UV spectral data suggest that hexabromocyclododecane does not absorb at wavelengths >290 nm(4) and, therefore, is not expected to be susceptible to direct photolysis by sunlight(SRC). Monitoring data from remote areas of the globe indicate that hexabromocyclododecane is subject to long-range environmental transport in the atmosphere(5,6). The majority of long-range environmental transport is likely to occur in aerosol form(5).

ENVIRONMENTAL: Average hexabromocyclododecane levels in mothers' milk from the Stockholm, Sweden area increased from 0.13 pmol/g lipid in 1980 to 0.60 pmol/g lipid in 2004, with the highest levels reported in 2001 and 2002, 0.83 and 0.93 pmol/g lipid, respectively. Levels in Norwegian breast milk samples ranged from 0.25-2 ng/g lipids over a study period of 1986-2001(1).

ENVIRONMENTAL: The National Food Administration in Sweden has analyzed HBCDD and other substances in mother's milk from Swedish women in two different studies. The first study detected HBCDD in 12 of 33 samples. The samples were taken from primiparous, age 19-40, living in Uppsala County, Sweden, two weeks after delivery. The limit of detection was /less than/ 15 pg HBCDD/g mothers milk (fresh weight). On a lipid weight basis, the mean and maximum concentrations were 0.45 and 2.4 ng/g fat, respectively(1).

ENVIRONMENTAL: ... the time trend of the stereoisomer-specific concentrations of HBCD (alpha, beta, and gamma) in breast milk samples of Japanese women by using liquid chromatography-tandem mass spectrometry (LC/MS/MS) was investigated. Time trend of summation operator HBCD appeared to be related to that of the industrial HBCD consumption level in Japan.

Human milk samples were collected from individuals residing in various regions across Canada ... in the years 1992 to 2005. These included five large cities in southern Canada as well as samples from Nunavik in northern Quebec. Comparative samples were also collected from residents of Austin, Texas, USA in 2002 and 2004. More than 300 milk samples were analysed for the brominated flame retardants (BFRs), PBDEs and HBCD, by extraction, purification and quantification using either isotope dilution gas chromatography-mass spectrometry (GC-MS) or liquid chromatography-MS. ...HBCD levels in most milk samples were usually less than 1ppb milk lipid and dominated by the a-isomer. This large data set of BFRs in Canadian human milk demonstrates an increase in the last few decades in human exposure to BFRs which now appears to have stabilized.

Toxicity Summary · 2건

IDENTIFICATION AND USE: Technical products of Hexabromocyclododecane (HBCDD) predominantly consist of three stereoisomers (alpha-, beta- and gamma-HBCDD). HBCDD is a widely used brominated flame retardant. HBCDD is used in several products, some of which are available to consumers. Some examples of end-products containing HBCDD are: flat and pile upholstered furniture, upholstery seatings in transportation, draperies, and wall coverings, bed mattress ticking, interior textiles, automobile interior textiles, car cushions, insulation boards of transport vehicles, insulation boards in building constructions, parking decks, insulation boards against frost heaves of road and railway embankments, packaging material, electrical and electronic equipment, video cassette housings, polyvinyl chloride wire, cable and textile coating, protecting paints for military purposes. HUMAN EXPOSURE AND TOXICITY: 10% HBCDD did not cause human skin sensitization. Although no enantioselective enrichment was detected in either dust or diet, substantial enrichment of (-)alpha-HBCDD was observed in human serum. Serum concentrations of HBCDD were correlated with the exposure via dust, but not via dietary ingestion. The enrichment of the (-)alpha-HBCDD enantiomer in humans appears to be due to in vivo enantioselective metabolism/excretion rather than ingestion of dust or diet. HBCDD display estrogenic effects in human cells in vitro. HBCDD modulates thyroid receptor-mediated gene expression in HeLaTR cells, but only in the presence of T3. HBCDD has been tested in the in vitro mammalian cytogenetic test using human peripheral blood lymphocytes, from a healthy 40 year adult female, both in the absence and presence of metabolic activation. No statistically significant increases in structural chromosome aberrations were observed in either the non-activated or activated studies, regardless of dose level or harvest time. Nor were there any increases in numerical chromosome aberrations. Cytotoxicities of HBCDD enantiomers were evaluated in Hep G2 cells. The order of cytotoxicity was gamma-HBCDD beta-HBCDD>alpha-HBCDD. In addition, HBCDD decreases the lytic function of human natural killer (NK) cells. ANIMAL STUDIES: Toxicological studies have been carried out using different experimental designs with single or repeated administration during gestation, postnatally or in adulthood using technical HBCDD. The composition of these mixtures differs from the HBCDD profile found in wildlife and in foods. Main targets for HBCDD toxicity were the liver, thyroid hormone homeostasis, the reproductive, the nervous and the immune systems. The activation of constitutive androstane receptor- (CAR) or pregnane-X-receptor (PXR)-dependent gene expression, leading to disruption of thyroid hormone homeostasis, is considered to be associated with neurodevelopmental effects on behavior and may also be responsible for effects on reproduction. The available studies indicate that HBCDDs are not genotoxic. There is limited information from only one long-term toxicity/carcinogenicity study for HBCDDs in mice, indicating that the incidence of altered foci in the liver of males was increased, as was the incidence of liver carcinoma in females, but without a dose-relationship. ECOTOXICITY STUDIES: Environmental study demonstrates that HBCDD affects breeding behavior in American kestrels throughout the reproductive season and behavioral alterations were linked to reproductive changes (egg size). These studies suggested that birds may be more sensitive to HBCDD than mammals. Other studies indicate limited potential for in vivo endocrine disruption of the reproductive and thyroid hormonal system in immature flounder. Juvenile rainbow trout experiments indicate that all HBCDD isomers have the potential to disrupt thyroid homeostasis. In the study the embryos of marine medaka (Oryzias melastigma) HBCDD exposure at environmentally realistic concentrations induced oxidative stress and apoptosis, and suppressed nucleotide and protein synthesis, which all together resulted in developmental toxicity, particularly in the cardiovascular system, in the embryos of O. melastigma. In plants, both the inhibitory effect of the diastereoisomers on the early development of maize and the intensities of hydroxyl radical and histone H2AX phosphorylation in maize exposed to HBCDD followed the order alpha-HBCD > beta-HBCD > gamma-HBCD, indicating the diastereomer-specific oxidative stress and DNA damage in maize.

The brominated flame retardants tetrabromobisphenol A (TBBPA) and hexabromocyclododecane (HBCD) are found in the environment, e.g., in sediments and organisms, in food items, human blood samples and mother's milk. In this study, the effects of both compounds on rat hepatic cytochrome P450 (CYP) levels and activities were investigated. Juvenile/young male and female Wistar rats were treated orally with various doses via the feed (TBBPA) or by gavage (HBCD). After 28 days of treatment the animals were sacrificed and hepatic mRNA and microsomes were isolated. HBCD treatment led to a significant induction of CYP2B1 mRNA, CYP2B1/2B2 protein and 7-pentoxyresorufin O-depentylase (PROD) activity suggesting a phenobarbital-type of induction. Furthermore, a significant increase in CYP3A1/3A3 mRNA, CYP3A1 protein, and luciferin benzylether debenzylase (LBD) activity was found, being more pronounced in females than in males. The effect on CYP3A1/3A3 mRNA was significant in female rats at a daily dose of 3.0 mg/kg bw and above. HBCD exhibited no effects on CYP1A2 mRNA, CYP1A1/1A2 protein, or microsomal 7-ethoxyresorufin O-deethylase (EROD) activity suggesting lack of activation of the aryl hydrocarbon receptor. No significant effects on any of the parameters measured were obtained with TBBPA. Our findings suggest that oral exposure to HBCD induces drug-metabolising enzymes in rats probably via the CAR/PXR signalling pathway. Induction of CYPs and co-regulated enzymes of phase II of drug metabolism may affect homeostasis of endogenous substrates including steroid and thyroid hormones.

Biological Half-Life · 5건

Although difficult to draw any conclusions on elimination based on so few time points, the data may perhaps suggest elimination half-life in the order of weeks to months for the three diastereomers, with the longest half-life for the alpha-diastereomer.

A single oral dose (7-9 mg/kg) of 14C-HBCD was administered to male (n=2) and female (n=8) rats. ...The estimated absorption half-life was 2 hours; peak radioactivity was detected in blood 4 hours post-dosing. ... Roughly 80 ->90% of the gamma stereoisomer was eliminated within 3 days following a single oral dose, with an apparent half-life of elimination of 27 hours.

The degradation of the enantiomers of a-, b-, and g-hexabromocyclododecane (HBCD) by phase I metabolism was investigated using induced rat liver microsomes. HBCD isomers were quantified using HPLC-MS/MS (ESI(-)) after separation on a combination of a reversed phase and a chiral analytical column. The degradation of all six isomers followed first-order kinetics and the estimated half-lives ranged from 6.3 min for both b-HBCD enantiomers to 32.3 min in case of (+)-g-HBCD. (+)-a- and (-)-g-HBCD displayed significantly shorter half-lives than their corresponding antipodes. It could be shown that this degradation led to a significant enrichment of the first eluting enantiomers (-)-a- and (+)-g-HBCD. Individual patterns of mono- and dihydroxylated derivatives obtained from each a- and g-HBCD enantiomer were seen to be distinctly characteristic. The patterns of monohydroxylated HBCD derivatives detected in liver and muscle tissues of pollack, mackerel and in herring gull eggs were largely similar to those observed in the in vitro experiments with rat liver microsomes. This enabled individual hydroxy-HBCDs to be assigned to their respective parent HBCD enantiomers.

Elimination /of a-HBCD following a single and repeated exposure with respect to dose, time, and route of administration in female C57BL/6 mice/, both whole-body and from individual tissues, was biphasic. a-HBCD-derived radioactivity was excreted in the feces as parent and metabolites, whereas urine only contained metabolites. Presence of polar metabolites in the blood and urine were a major factor in determining the rapid initial whole-body half-life after a single oral exposure. Initial half-lives were approximately1-3 days and much longer terminal half-lives of 17 days were observed, suggesting the potential for a-HBCD bioaccumulation.

/In mice/ polar metabolites in the blood and urine were a major factor in determining the initial whole-body half-life (1 day) after a single po exposure. Elimination, both whole-body and from individual tissues, was biphasic. Initial half-lives were approximately 1 day, whereas terminal half-lives were up to 4 days, suggesting limited potential for g-diastereoisomer bioaccumulation.

HBCD was detected in the adipose tissue of male and female rats treated with 1000 mg/kg/day for up to 90 days. Isomer-specific analysis showed that the relative isomer concentrations in adipose tissue at all time points were alpha>>gamma>beta which is in contrast to the test article composition (gamma>>alpha>beta). Steady state levels were achieved by study day 27. Levels in male and female rats were similar at all time points and declined during the recovery period.

Juvenile rainbow trout (Oncorhynchus mykiss) were exposed to three diastereoisomers (alpha, Beta, gamma) of hexabromocyclododecane (C12H18Br6) via their diet for 56 days followed by 112 days of untreated food to examine bioaccumulation parameters and test the hypothesis of in vivo bioisomerization. Four groups of 70 fish were used in the study. Three groups were exposed to food fortified with known concentrations of an individual diastereoisomer, while a fourth group were fed unfortified food. Bioaccumulation of the gamma-diastereoisomer was linear during the uptake phase, while the alpha- and Beta-diastereoisomers were found to increase exponentially with respective doubling times of 8.2 and 17.1 days. Both the Beta- and the gamma-diastereoisomers followed a first-order depuration kinetics with calculated half-lives of 157 + or - 71 and 144 + or - 60 days (+ or -1 x standard error), respectively. The biomagnification factor (BMF) for the alpha-diastereoisomer (BMF = 9.2) was two times greater than the Beta-diastereoisomer (BMF = 4.3); the large BMF for the alpha-diastereoisomer is consistent with this diastereoisomer dominating higher-trophic-level organisms. Although the BMF of the Beta-diastereoisomer suggests that it will biomagnify, it is rarely detected in environmental samples because it is present in small quantities in commercial mixtures. Results from these studies also provide evidence of bioisomerization of the Beta- and gamma-diastereoisomers. Most importantly, the alpha-diastereoisomer that was recalcitrant to bioisomerization by juvenile rainbow trout in this study and known to be the dominant diastereosiomer in fish was bioformed from both the Beta- and the gamma-diastereoisomers.

A single oral dose (7-9 mg/kg) of 14C-HBCD was administered to male (n=2) and female (n=8) rats. ...Only metabolites were detected in feces and urine -no parent molecule was detected. Thus, the gamma isomer appears to be extensively metabolized prior to excretion. In contrast, only unmetabolized gamma isomer was detected in adipose tissue.

The laboratory trophic transfer of hexabromocyclododecanes (HBCDs) was studied using predatory (oscar) fish and a prey species (tiger barb) exposed to a technical HBCD. Gut absorption, dynamic changes of diastereomer pattern and enantiomer fractions, and potential metabolism of HBCDs were examined. Compared with b- or g-HBCD, a-HBCD showed lower absorption efficiency in the gut of oscar fish. A predominance of g-HBCD was observed in the tiger barb after 5 d HBCD-exposed and oscar feeding on the tiger barb for 16 d. After 20 d of depuration, 41.1% g-HBCD and 42.7% b-HBCD disappeared, and a-HBCD exceeded the initial amount. The transformation from g-HBCD predominance in the food to a-HBCD predominance in the oscar was attributed mainly to the isomerization of g-HBCD (at least 3% and up to 22.7%) to a-HBCD. Selective enrichment of the (+) a- and (-) b-enantiomers and no enantioselective enrichment of g-HBCD were observed in the tiger barbs. No enantioselective uptake of the 3 diasteromers was found in the oscar gut. The enantiomer fractions of a- and g-diastereomers were significantly higher, but that of b-diastereomer were significantly lower in the oscars than in the tiger barbs, indicating enantioselective metabolism of the 3 diastereomers. Two HBCD monohydroxylated metabolites were detected in the 2 fish species, but their composition patterns differed, indicating a species-specific metabolism of HBCD in the studied fish species.

Bromine is mainly absorbed via inhalation, but may also enter the body through dermal contact. Bromine salts can be ingested. Due to its reactivity, bromine quickly forms bromide and may be deposited in the tissues, displacing other halogens. (L626)

Absorption, Distribution and Excretion · 5건

...Rats administered a single oral dose of 1.93 mg of radiolabeled HBCD eliminated 86% of the dose within 72 hr. (The total dose administered was 7-9 mg/kg body weight.) Absorption from the gastrointestinal tract reportedly occurred rapidly, with a half-life of 2 hr. However, the amount of the absorbed fraction was not reported. HBCD was reported to be rapidly metabolized and eliminated in the feces and urine following absorption, with 70% of the administered radioactivity eliminated in the feces and another 16% eliminated in the urine 72 hr after dosing. A two-compartment model was constructed, with non-adipose tissues in one compartment and adipose tissue in the other. Elimination from the adipose compartment was reported to be slower than elimination from the non-adipose compartment, although elimination half-times were not provided in the review. In another study, ... HBCD was orally administered to male Wistar rats (number not reported) in olive oil at 500 mg/kg/day for 5 days. HBCD was found to be present only in adipose tissue, and in none of the other organs examined (i.e., spleen, pancreas, liver, kidneys, and heart). HBCD was found to be excreted in the feces, with an average of 32-35% of the cumulative administered dose excreted. No HBCD was found in the urine. Although differences in study design, including the test vehicle and the analytic methods used, may account for some of the difference in the results, both studies... suggest that following acute oral doses, HBCD is rapidly absorbed from the gastrointestinal tract, distributed primarily to body fat, and eliminated rapidly, primarily in the feces.

The excretion of HBCD in urine and feces as well as its distribution to various organs was investigated. The test article was "Pyroguard SR-103"... . A fine suspension of SR-103 in olive oil was prepared by mixing well to homogeneity in a mortar. The dose was 500 mg/kg/day for 5 consecutive days (n= 4 male Wistar rats). ...The average daily rate of excretion in the feces was 29-37% of the dose. The cumulative excretion was roughly constant at 32-35% with respect to the cumulative administered amount. Urinary excretion was not observed. No evidence for the presence of metabolites was observed in urine or feces. A separate study with isolated intestinal loop (upper jejunum) indicated about 12% of the dose was detectable in the intestinal tissue and the amount remaining in the lumen loop "small". The test article was detected only in the adipose tissue after dosing for 5 days. The level in adipose tissue was 0.3-0.7 mg/g fat.

A single oral dose (7-9 mg/kg) of 14C-HBCD was administered to male (n=2) and female (n=8) rats. Based on the starting material and the melting point of the final product described in the report, the test article appeared to be composed of the gamma isomer. The rats were sacrificed 8, 24, 48 and 72 hours (females) and 48 hours (males) after dosing. One female rat served as control. Urine and feces were collected daily, blood samples from 4 animals were collected during the first 24 hours. Tissue samples were collected at the time of sacrifice. ... HBCD appeared to be well absorbed from the gastrointestinal tract and extensively metabolized prior to elimination in feces (primary route) and urine. The estimated absorption half-life was 2 hours; peak radioactivity was detected in blood 4 hours post-dosing. The pharmacokinetics of the gamma stereoisomer appeared to follow an open two-compartment model. The central compartment was described as liver, lung, kidney, heart, muscle, gonads, uterus, spleen, and brain; the peripheral compartment was described as adipose tissue. Roughly 80 ->90% of the gamma stereoisomer was eliminated within 3 days following a single oral dose, with an apparent half-life of elimination of 27 hours. Only metabolites were detected in feces and urine -no parent molecule was detected. Thus, the gamma isomer appears to be extensively metabolized prior to excretion. In contrast, only unmetabolized gamma isomer was detected in adipose tissue. In females at 8, 24, 48 and 72 hours post-dosing, the total 14C activity detected in tissues of female rats was about 43, 24, 18 and 17% of the dose, respectively. In male rats at 48 hours post-dosing (the only time point investigated in males), the 14C-activity was about 10% of the dose. Similarly, that detected in feces from female rats was about 4, 65, 54 and 77% at 8, 24, 48 and 72 hr. Urine contained 0.1, 6, 18 and 15% of the dose at the same time intervals. Feces from male rats at 48 hours contained about 94% of the dose while urine from male rats contained about 15%. At 48 hours post-dosing approximately 81% of the dose was detected in feces and urine of female rats. Thus, at 48 hours post-dosing approximately 86% of the dose was recovered in the tissues, feces and urine from female rats whereas 119% was recovered from males. The lower 48 hour recovery from this group of female rats is largely accounted for by the lower fecal content (43%) of radioactivity collected during the 0-24 hours post-dosing. By 48 hours post-dosing, females sacrificed at 24 hours, had a total fecal 14C-content of 65% of the dose, and females sacrificed at 72 hours had a total fecal 14C-content of 62% of the dose. Substituting the average value, 63%, from these two groups for the 43% value used to calculate overall recovery, the total per cent of the dose accounted for at 48 hours becomes feces (74%) urine (17%) and tissues (18%) or 109% of the dose. It appears likely that some unknown factor resulted in the lower 0-24 hour percent recovery from feces, and that percent of dose present in tissues, feces and urine 48 hours post-dosing is similar in male and female rats.

Seven week old Wistar rats were administered HBCDD by gavage for 28 days. The HBCDD was properly dissolved in corn oil ... The composition of diastereomers was 10.3 - 8.72 - 81.0% of alpha, beta, and gamma, respectively ... The doses were 0, 0.3, 1, 3, 10, 30, 100, and 200 mg/kg bw/day ... The chemical analysis showed that nearly no beta-HBCDD was present in the animals. The concentration of alpha-HBCDD and gamma-HBCDD in liver fat increased dose-dependently, but there were signs of a plateau being reached at the highest dose level(s), and perhaps earlier for gamma than for alpha. Accordingly, with increasing dose, more and more of the HBCDD-residues consisted of the alpha diastereomer. Females consistently accumulated more HCBDD in the liver compared to males, but as no absolute concentrations are presented, it is difficult to quantify the difference. At doses up to the levels where the residue levels seem to plateau (30-200 mg/kg/day), the accumulation of alpha seemed to be roughly twice as high as gamma. At doses of 100 mg/kg/day and above, the relative concentration of alpha increased much further compared to gamma. This may be explained by either reduced absorption or increased metabolism of gamma at high doses. The latter is possibly explained by induction of CYP 2B.

An aliphatic, brominated cyclic alkane produced by bromination of cyclododecatri-1,5,9-ene

Technical grade HBCDD is generally produced from cis, trans, trans-1,5,9-cyclododecatriene (CDT), one of four CDT isomers, (CAS No. 27070-59-3). The reaction, trans-addition of bromine to the double bonds of CDT, results in the three diastereomers alpha-, beta- and gamma- HBCDD.

... Produced by the addition reaction of bromine to trans, trans, cis-1,5,9-cyclododecatriene, the cyclic trimer of butadiene. The hexabromocyclododecane that is obtained from this reaction consists of a mixture of stereoisomers of 1,2,5,6,9,10-hexabromocyclododecane ranging from oils to an isomer with a 205-208 °C melting point. Commercially available hexabromocyclododecane contains a mixture of solid isomers and has a melting point range of 170-180 °C (185-195 °C).

The final distribution of the diastereomers in technical HBCDD varies with a range of about 70-95% gamma-HBCDD and 5-30 % alpha- and beta-HBCDD.

Technical HBCDD is manufactured in two forms, high-melting (HM) and low-melting (LM). The low-melting HBCDD consists of 70-80% gamma-, 20-30% of alpha- and beta-HBCDD. The high-melting HBCDD consists of 90% or more of gamma-HBCDD.

According to Material Safety Data Sheets from Great Lakes /Chemical Corporation/ there are three technical products of hexabromocyclododecane. One product is 100% pure, another product contains an inorganic stabilizer, and a third product contains 40-60% HBCDD, water and a component A. The Dead Sea Bromine Group has two technical products, one which is 99.5% pure and another which is heat stabilized grade. Albemarle also has two technical products, one of high-purity grade available in powder or granular form, the other one of high-purity grade is ground to a fine particle size.

Technical products of HBCDDs (containing 73-74 % Br) predominantly consists of three stereoisomers (alpha-, beta- and gamma-HBCDD) present in relative amounts of 9-13 % alpha-HBCDD, <0.5-12 % beta-HBCDD and 72-90 % gamma-HBCDD. Apart from these three, also delta- and epsilon-HBCDD may be present in the technical product, although at very low concentrations. In addition, the commercial HBCDDs may contain 1-2 % tetrabromocyclododecene.

용도The commercially available brominated flame retardant hexabromocyclododecane (HBCD) is lipophilic, has a high affinity to particulate matter and low water solubility. It is used as a flame retardant additive, with the intent of delaying ignition and slowing subsequent fire growth during the service life of vehicles, buildings or articles, as well as while materials are stored. The main uses of HBCD are in flame-retarded expanded (EPS) and extruded (XPS) polystyrene foam insulation, with smaller scale use in textile applications and electric and electronic appliances (high impact polystyrene/HIPS). In textiles HBCD is used in back-coatings for upholstery and other interior textiles, including automotive applications.

The aliphatic flame retardant, hexabromocyclododecane (HBCD), is produced by the addition reaction of bromine to trans,trans,cis-1,5,9-cyclododecatriene, the cyclic trimer of butadiene. The HBCD that is obtained from this reaction consists of a mixture of stereoisomers of 1,2,5,6,9,10-hexabromocyclo-dodecane [3194-55-6] ranging from oils to an isomer with a 205–208℃ melting point. Commercially available HBCD contains a mixture of solid isomers and has a melting point range of 170–180 ℃ (185–195℃ ). Addition of bromine to the cyclic triene in nonpolar solvents results in substantial amounts of free-radical substitution reactions which lead to decreased yields and lower quality product. Use of polar solvents, such as ethanol, reduces the substitution reaction but can give rise to vicinal bromoethoxy by products. Mixed solvent systems, such as benzene–tert-butyl alcohol or perchloroethylene-ethanol,can be employed to improve product purities and yields. Another technique to reduce the formation of undesired byproducts is to add the bromine and cyclododecatriene simultaneously and sep- arately to ethanol. This process is carried out by separately feeding bromine and cyclo- dodecatriene in a molar ratio of 3 : 1 to 96 % ethanolinastirredreactor. The reaction temperature is kept at 20-30℃ and a 1-5% excess of bromine is maintained during the 2.5–3.0h addition. The precipitated product is recovered by filtration or centrifugation, and the mother liquor is recovered and reused in subsequent reactions after adding makeup ethanol. The total yield of dried HBCD from nine separate batches using recycled mother liquor is 92%.

Hepatocellular carcinoma (HCC) is one of the most common cancers in China with high mortality, high chemotherapy resistance incidence, and poor prognosis. This study aimed to investigate the influence of polychlorinated biphenyls (PCBs) and hexabromocyclododecane (HBCD) on chemoresistance of HCC cells (HepG2, MHCC97H, and MHCC97L) to cisplatin and to explore the potential molecular mechanism. Cell viability, DNA damage, the expression level and activity of nuclear factor-kB (NF-kB), p53/Mdm4, and phosphatidylinositol 3-kinase/protein kinase B (PI3K/AKT) pathway were measured. The results showed that HBCD and PCBs could significantly reduce the chemosensitivity of HCC cells to cisplatin, increasing the cell viability and decreasing DNA damage. Moreover, HBCD and PCBs could induce the transcriptional activity of NF-kb and suppress the p53 expression in HepG2 and MHCC97H cells. In MHCC97L cells, however, opposite changes for NF-kB protein expression, NF-kB transcriptional activity, and p53/Mdm4 expression were observed after HBCD and PCBs exposure. Further investigation revealed that HBCD and PCBs exposure significantly increased the expression level of p-Akt and mammalian target of rapamycin (mTOR) in HepG2 and MHCC97H cells, but reduced that in MHCC97L cells. PI3K inhibitor LY294002 could relieve the influence of HBCD and PCBs on chemoresistance in HepG2 and MHCC97H cells. Taken together, HBCD and PCBs at low concentrations could increase the resistance of HCC cells to cisplatin through modulation on NF-kB pathway activation and p53 function, which is associated with the activity of PI3K/Akt pathway.

Bromine is a powerful oxidizing agent and is able to release oxygen free radicals from the water in mucous membranes. These free radicals are also potent oxidizers and produce tissue damage. In additon, the formation of hydrobromic and bromic acids will result in secondary irritation. The bromide ion is also known to affect the central nervous system, causing bromism. This is believed to be a result of bromide ions substituting for chloride ions in the in actions of neurotransmitters and transport systems, thus affecting numerous synaptic processes.
