Mechanism of Action · 1건

The acquisition of steroidogenic competence by prothoracic glands of last instar Manduca sexta larvae is regulated by juvenile hormone (JH). Topical treatment of pre-commitment larvae with JH I or (7S)-hydroprene (a JH analog) delays development by increasing the time to pupal commitment and wandering. Prothoracic gland competence is suppressed in JH-treated larvae: Unstimulated and prothoracicotropic hormone (PTTH)-stimulated rates of in vitro ecdysone secretion are decreased relative to rates of secretion by competent glands. (7S)-Hydroprene also suppresses the competence of glands in head-ligated pre-commitment larvae, suggesting the hormone acts directly on the glands. Two results indicate PTTH plays a role in controlling competence, and that JH regulates competence indirectly by inhibiting PTTH release: (1) head-ligation prevents the acquisition of full competence, and (2) cAMP levels are elevated in glands from JH-treated larvae. Thus, the decrease in the JH titer that precedes pupal commitment in Manduca is permissive for the acquisition by prothoracic glands of steroidogenic competence.

Environmental Fate · 3건

TERRESTRIAL FATE: Based on a classification scheme(1), an estimated Koc value of 9320(SRC), determined from a structure estimation method(2), indicates that S-hydroprene is expected to be immobile in soil(SRC). Volatilization of S-hydroprene from moist soil surfaces is expected to be an important fate process(SRC) given an estimated Henry's Law constant of 4.2X10-5 atm-cu m/mole(SRC), based upon its vapor pressure, 3X10-4 mm Hg(3), and water solubility, 2.5 mg/L(3). However, adsorption to soil is expected to attenuate volatilization(SRC). S-Hydroprene is not expected to volatilize from dry soil surfaces(SRC) based upon its vapor pressure(3). Biodegradation data in soil were not available(SRC, 2016).

AQUATIC FATE: Based on a classification scheme(1), an estimated Koc value of 9320(SRC), determined from a structure estimation method(2), indicates that S-hydroprene is expected to adsorb to suspended solids and sediment(SRC). Volatilization from water surfaces is expected(3) based upon an estimated Henry's Law constant of 4.2X10-5 atm-cu m/mole(SRC), derived from its vapor pressure, 3X10-4 mm Hg(4), and water solubility, 2.5 mg/L(4). Using this Henry's Law constant and an estimation method(3), volatilization half-lives for a model river and model lake are 39 hours and 17 days, respectively(SRC). However, volatilization from water surfaces is expected to be attenuated by adsorption to suspended solids and sediment in the water column. The estimated volatilization half-life from a model pond is 19 months if adsorption is considered(5). S-Hydroprene is not expected to undergo hydrolysis in the environment due to the lack of functional groups that hydrolyze under environmental conditions(3). According to a classification scheme(6), an estimated BCF of 9030(SRC), from its log Kow of 6.5(4) and a regression-derived equation(2), suggests the potential for bioconcentration in aquatic organisms is very high, provided the compound is not metabolized by the organism(SRC). Biodegradation data in water were not available(SRC, 2016).

ATMOSPHERIC FATE: According to a model of gas/particle partitioning of semivolatile organic compounds in the atmosphere(1), S-hydroprene, which has a vapor pressure of 3X10-4 mm Hg at 25 °C(2), is expected to exist solely as a vapor in the ambient atmosphere. Vapor-phase S-hydroprene is degraded in the atmosphere by reaction with photochemically-produced hydroxyl radicals and ozone(SRC); the half-lives for these reactions in air are estimated to be 4.8 hours and 48 minutes(SRC), calculated from its respective rate constants of 8X10-11 and 3.4X10-16 cu cm/molecule-sec at 25 °C(SRC) that were derived using a structure estimation method(3). S-Hydroprene contains chromophores that absorb at wavelengths >290 nm(4) and, therefore, may be susceptible to direct photolysis by sunlight(SRC).

Toxicity Summary · 1건

IDENTIFICATION AND USE: S-Hydroprene is an amber liquid (technical). It is an insect growth regulator. It inhibits normal molting processes causing mortality or sterility of insect at or before the final molt. HUMAN EXPOSURE AND TOXICITY: There are no data available. ANIMAL STUDIES: In a 13-week rat study using S-hydroprene, there were disturbances in hematological and clinical chemistry parameters, including prothrombin time (dose-related decrease in males). Histopathologically, there was increased homogeneity of liver cytoplasm at high doses and in females, vacuolated interstitial cells in the ovaries. S-hydroprene was not carcinogenic in rats. In a multigeneration study in rats, using S-hydroprene, there was some evidence of an effect on seminiferous tubules in males, associated with an effect on fertility; it was unclear whether these effects were test-material related. S-hydroprene was not fetotoxic or teratogenic in rat. S-hydroprene was not mutagenic.

/OTHER TERRESTRIAL SPECIES/ Results of the histomorphological studies of the ovaries in normal Achoea janata are compared to the effects induced by the juvenoid, S-hydropene (312-006) on the ovaries. S-hydropene elicits various derangemental changes on the ovary when administered to O-day old pupae of Achoea janata S-hydroprene treatment also resulted in resorption of oocytes in the vitellarium and reduced the number of oocytes. Thus the treatment of S-hydropene indicates the possibility of reducing the reproductive potential of the female moth.

Volatilization from Water/Soil · 1건

The Henry's Law constant for S-hydroprene is estimated as 4.2X10-5 atm-cu m/mole(SRC) derived from its vapor pressure, 3.0X10-4 mm Hg(1), and water solubility, 2.5 mg/L(1). This Henry's Law constant indicates that S-hydroprene is expected to volatilize from water surfaces(2). Based on this Henry's Law constant, the volatilization half-life from a model river (1 m deep, flowing 1 m/sec, wind velocity of 3 m/sec)(2) is estimated as 39 hours(SRC). The volatilization half-life from a model lake (1 m deep, flowing 0.05 m/sec, wind velocity of 0.5 m/sec)(2) is estimated as 17 days(SRC). However, volatilization from water surfaces is expected to be attenuated by adsorption to suspended solids and sediment in the water column. The estimated volatilization half-life from a model pond is 19 months if adsorption is considered(3). S-Hydroprene's estimated Henry's Law constant indicates that volatilization from moist soil surfaces may occur(SRC). S-Hydroprene is not expected to volatilize from dry soil surfaces(SRC) based upon its vapor pressure(1).
