Mechanism of Action · 5건

The peroxidative activity of human lung microsomes was shown to activate MOCA ((14)C-MOCA, specific activity 46.7 mCi/mmol) to bind DNA. With specific antisera it was demonstrated that myeloperoxidase was responsible for DNA binding. Binding was dependent on H2O2 and was inhibited by azide, but not by indomethacin or eicosatetraynoic acid.

The activation of carcinogenic aromatic and heterocyclic amines and benzo[a]pyrene-7,8-diol to intracellular electrophiles by prostaglandin H synthase (COX) is well documented for ovine sources of this enzyme. Here, the arachidonic acid-dependent activation of substrates by human (h)COX-1 and-2 is examined, utilizing recombinant enzymes. The COX-dependent activation of benzidine (BZ), 4-aminobiphenyl, (+)benzo[a]pyrene-7,8-diol, (+)benzo[a]pyrene-7,8-diol, 2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline (MeIQx), 2-amino-3-methylimidazo [4,5-f]quinoline (IQ), 2-amino-1-methyl-6-phenylimidazo [4,5-b] pyridine (PhIP), and 4,4'-methylenebis(2-chloroaniline) (MOCA) is assessed by means of COX-catalyzed, covalent DNA binding. The hCOX isozymes activated all substrates tested, activation varied from barely detectable for IQ (0.76 and 1.52 pmol bound/mg DNA for COX-1 and -2, respectively) to a high of 65 and 117 pmol bound/mg DNA for COX-1 and -2, respectively, for the activation of MOCA. BZ, which is an excellent peroxidase substrate, did not exhibit high DNA binding levels in hCOX assays and this phenomenon was found to be due to high levels of binding to protein, which effectively competed with the DNA for binding in the assay. The demonstrated ability of the COX enzymes to activate a variety of environmental and dietary carcinogens indicates a potential role for COX in the activation pathway of aromatic and heterocyclic amines and polycyclic hydrocarbons at extra-hepatic sites during early or late stages of carcinogenesis.

The mechanism of carcinogenesis is believed to involve the formation of chemical adducts in genetic material through covalent binding of electrophilic metabolites form MOCA to initiate carcinogenesis. ...Besides initiation, there is some evidence that MOCA may have tumorigenesis-promoting activity, as suggested by its ability to inhibit gap-junctional cell communication, which is important in controlling cell proliferation.

The mechanism of hemoglobin adduct formation by N-oxidized MBOCA metabolites was recently investigated in viva and in vitro using human and rat hemoglobin. Male Sprague-Dawley rats were injected ip with 0.5-50 mg/kg of MBOCA, and male English short hair guinea pigs were injected sc with 4-100 mg/kg of MBOCA. In the same study, treatment of rats with either phenobarbital or beta-naphthaloflavone (BnF) resulted in induction of cytochrome P-450 as evidenced by increased enzymatic activity of two P-450 isozymes. This induction caused increased ortho-hydroxylation of MBOCA that was not statistically significant and resulted in the formation of various metabolites. Two of those MBOCA metabolites, n-hydroxy MBOCA and mononitroso-MBOCA, formed measurable amounts of adducts with both human and rat hemoglobin, while the amount of adducts formed by the parent compound itself was very small. This in vitro binding occurred in a dose-related manner and could by inhibited by cysteine and glutathione but not by oxidized glutathione or methionine. Therefore, the administration of MBOCA to rats resulted in a dose-related formation of hemoglobin adducts that were detectable 24 days after treatment. Similar observations were made in guinea pigs; the life span of MBOCA-hemoglobin adducts followed that of re cells in the guinea pig and was approximately 60-80 days.

4,4'-Methylenebis

Causes hematuria, bladder cancer, and methemoglobinemia in animal toxicology studies; Listed in the table Examples of Industrial Chemicals for Which Methemoglobin Formation is NOT the Principal Cause of Toxicity; MBOCA has been clearly carcinogenic in rats, mice, and dogs after oral administration. In rats, lung and liver tumors were the most prominent. In mice, an increased incidence of hepatomas was seen in females. In dogs, bladder tumors were observed. . . . Hematuria has been reported in a small group of workers exposed to MBOCA together with a variety of other materials at unmeasured concentrations, e.g., toluene diisocyanate, polyester resins, and polyether resins. The urinary conditions were mild and cleared within a week. . . . ACGIH encourages implementation of a urine monitoring program to ensure the effectiveness of dermal exposure controls . . . . Data indicative of bladder cancer in workers exposed to MBOCA were inconclusive. [ACGIH] In most cases, dermal absorption after contact with contaminated surfaces is the most important occupational exposure route, with inhalation and ingestion representing minor exposure pathways. . . . The genotoxicity of 4,4'-methylenebis(2-chlorobenzenamine) is well documented and its toxicological profile is similar to that of orthotoluidine, thus indicating a common mode of action. 4,4'-methylenebis(2-chlorobenzenamine) has been shown to interact with DNA to form adducts in urothelial cells, and with haemoglobin to form adducts in the blood of workers exposed to this compound. . . . No adequate epidemiological studies were available to the Working Group to evaluate an association between MOCA and bladder-cancer risk. [Reference #1] Listed in Table 1 (Group 1 agents evaluated in the IARC Monographs Volumes 1-120, excluded from primary list of occupational carcinogens). Reason for exclusion: Group 1 classification based on mechanistic upgrade. [PMID 29769352]

Disposal Methods · 5건

Generators of waste (equal to or greater than 100 kg/mo) containing this contaminant, EPA hazardous waste number U158, must conform with USEPA regulations in storage, transportation, treatment and disposal of waste.

SRP: Wastewater from contaminant suppression, cleaning of protective clothing/equipment, or contaminated sites should be contained and evaluated for subject chemical or decomposition product concentrations. Concentrations shall be lower than applicable environmental discharge or disposal criteria. Alternatively, pretreatment and/or discharge to a permitted wastewater treatment facility is acceptable only after review by the governing authority and assurance that "pass through" violations will not occur. Due consideration shall be given to remediation worker exposure (inhalation, dermal and ingestion) as well as fate during treatment, transfer and disposal. If it is not practicable to manage the chemical in this fashion, it must be evaluated in accordance with EPA 40 CFR Part 261, specifically Subpart B, in order to determine the appropriate local, state and federal requirements for disposal.

PRECAUTIONS FOR "CARCINOGENS": There is no universal method of disposal that has been proved satisfactory for all carcinogenic compounds & specific methods of chem destruction ... published have not been tested on all kinds of carcinogen-containing waste. ... summary of avail methods & recommendations ... /given/ must be treated as guide only. /Chemical Carcinogens/

Spillage Disposal · 1건

Personal protection: complete protective clothing including self-contained breathing apparatus. Sweep spilled substance into covered containers. If appropriate, moisten first to prevent dusting. Carefully collect remainder. Then store and dispose of according to local regulations. Do NOT let this chemical enter the environment.

Preventive Measures · 5건

SRP: Contaminated protective clothing should be segregated in such a manner so that there is no direct personal contact by personnel who handle, dispose, or clean the clothing. The completeness of the cleaning procedures should be considered before the decontaminated protective clothing is returned for reuse by the workers. Contaminated clothing should not be taken home at the end of shift, but should remain at employee's place of work for cleaning.

SRP: Local exhaust ventilation should be applied wherever there is an incidence of point source emissions or dispersion of regulated contaminants in the work area. Ventilation control of the contaminant as close to its point of generation is both the most economical and safest method to minimize personnel exposure to airborne contaminants. Ensure that the local ventilation moves the contaminant away from the worker.

PRECAUTIONS FOR "CARCINOGENS": Smoking, drinking, eating, storage of food or of food & beverage containers or utensils, & the application of cosmetics should be prohibited in any laboratory. All personnel should remove gloves, if worn, after completion of procedures in which carcinogens have been used. They should ... wash ... hands, preferably using dispensers of liq detergent, & rinse ... thoroughly. Consideration should be given to appropriate methods for cleaning the skin, depending on nature of the contaminant. No standard procedure can be recommended, but the use of organic solvents should be avoided. Safety pipettes should be used for all pipetting. /Chemical Carcinogens/

PRECAUTIONS FOR "CARCINOGENS": In animal laboratory, personnel should remove their outdoor clothes & wear protective suits (preferably disposable, one-piece & close-fitting at ankles & wrists), gloves, hair covering & overshoes. ... clothing should be changed daily but ... discarded immediately if obvious contamination occurs ... /also,/ workers should shower immediately. In chemical laboratory, gloves & gowns should always be worn ... however, gloves should not be assumed to provide full protection. Carefully fitted masks or respirators may be necessary when working with particulates or gases, & disposable plastic aprons might provide addnl protection. If gowns are of distinctive color, this is a reminder that they should not be worn outside of lab. /Chemical Carcinogens/

Urinary concentrations of 70-1500 ug/L were found in samples from workers in one area of a 4,4'-methylenebis(2-chloroaniline) production facility(1). Between 1980 and 1983, 16.9% of 3,323 urinary samples submitted by 54 US companies contained 4,4'-methylenebis(2-chloroaniline) at concentrations >50 ug/L(1). Process workers at a polyurethane production unit were found to have up to 38 umol/mol creatinine of 4,4'-methylenebis(2-chloroaniline) in their urine at the end of the workshift(2). Smaller quantities were found in the urine of laboratory and supervisory staff, but none was detected in the urine of office and sales staff from an unrelated industry(2). At a commercial manufacturing plant that manufactures polyurethane products, the concentration of 4,4'-methylenebis(2-chloroaniline) in the urine of molders and mixers averaged 28.8 ug/L with maximum concentration of 158.9 ug/L(3). At a polyurethane elastomer factory, the mean urinary 4,4'-methylenebis(2-chloroaniline) levels for works at the beginning and end of the work shift ranged from 3.1-81.5 and 2.4-96.6 ug/g creatinine, respectively(4).

At a commercial manufacturing plant that manufactures polyurethane products, 4,4'-methylenebis(2-chloroaniline) was detected at <0.015 to 0.70 ug/filter(1). At a polyurethane elastomer factory, personal air exposure levels of 4,4'-methylenebis(2-chloroaniline) ranged from 0.2 to 8.9 ug/cu m(2).

MBOCA metabolism is influenced by phenobarbital. In vivo treatment with phenobarbital induced cytochrome P-450 enzymes which resulted in a slight increase in MBOCA hydroxylation. Rats treated with phenobarbital had a 4-8 fold increased metabolic rate for MBOCA. Phenobarbital did not, however, affect adduct formation.

In vivo treatment with P-naphthoflavone did increase the rate of MBOCA-hemoglobin adduct formation in rats treated sc with either 100 or 500 mg/kg/day.

MBOCA metabolism is NADPH-dependent. Moreover, MBOCA hydroxylation is inhibited by 2,4-dichloro-6-phenylphenoxyethylamine, an inhibitor of microsomal mixed function oxidases.

Cysteine and glutathione inhibit invivo hemoglobin adduct formation by N-hydroxy-MBOCA and mononitroso-MBOCA. The most recent findings suggest that binding of N-hydroxy-MBOCA to DNA in rat tissues can be inhibited by ascorbic acid, glutathione, nitrosobenzene, and methyl viologen but not by nitromethane, p-nitrobenzylpyridine, or methionine.

A 0% of theoretical BOD using activated sludge in the Japanese MITI test(5) suggests that biodegradation is not an important environmental fate process in soil(SRC).

AQUATIC FATE: Based on a classification scheme(1), an estimated Koc value of 5700(SRC), determined from a structure estimation method(2), indicates that 4,4'-methylenebis(2-chloroaniline) is expected to adsorb to suspended solids and sediment(SRC). Volatilization from water surfaces is not expected(3) based upon an estimated Henry's Law constant of 1.1X10-11 atm-cu m/mole(SRC), developed using a fragment constant estimation method(4). According to a classification scheme(5), BCFs of 114 to 398 in carp(6), suggest bioconcentration in aquatic organisms is high(SRC). Hydrolysis is not expected to be important for 4,4'-methylenebis(2-chloroaniline) based on a hydrolysis half-life of >800 years at pH 7 and 25 °C(7). A 0% of theoretical BOD using activated sludge in the Japanese MITI test(6) suggests that biodegradation is not an important environmental fate process in water(SRC).

ATMOSPHERIC FATE: According to a model of gas/particle partitioning of semivolatile organic compounds in the atmosphere(1), 4,4'-methylenebis(2-chloroaniline), which has an estimated vapor pressure of 3.9X10-6 mm Hg at 25 °C(SRC), determined from a fragment constant method(2), will exist in both the vapor and particulate phases in the ambient atmosphere. Vapor-phase 4,4'-methylenebis(2-chloroaniline) is degraded in the atmosphere by reaction with photochemically-produced hydroxyl radicals(SRC); the half-life for this reaction in air is estimated to be 5.0 hours(SRC), calculated from its rate constant of 7.8X10-11 cu cm/molecule-sec at 25 °C(SRC) that was derived using a structure estimation method(3). Particulate-phase 4,4'-methylenebis(2-chloroaniline) may be removed from the air by wet or dry deposition(SRC). 4,4'-Methylenebis(2-chloroaniline) contains chromophores that absorb at wavelengths >290 nm(4) and, therefore, may be susceptible to direct photolysis by sunlight(SRC).

Induction of hepatic ornithine-decarboxylase (ODC) activity by 4,4'-methylene-bis(2-chloroaniline) (MOCA) was studied in Male Sprague-Dawley-rats. ...Maximum induction of ODC activity after a single ip dose of 75 mg/kg MOCA occurred 18 hours after injection; activity returned to the control value 42 hours after dosing. In rats that received two 75 mg/kg doses, the extent of ODC induction after the second dose was similar to that of the first dose. MOCA at 15 mg/kg did not significantly increase ODC activity. Oral induction of ODC activity by 75 mg/kg MOCA was less than ip induction but significantly greater than the control value at 18 hours.

Metabolism/Metabolites · 6건

Although little is known about the disposition of MBOCA, it is likely that it is metabolized via the pathways that have been demonstrated for other aromatic amines. These pathways include N-oxidation, N-acetylation, C-oxidation, and conjugation with glucuronate or sulfate.

Arylamine N-acetyltransferases (NATs; EC 2.3.1.5) catalyze both the N-acetylation and O-acetylation of arylamines and N-hydroxyarylamines. ... Recombinant rat Nat3 was functional and catalyzed the N-acetylation of several arylamine substrates, including ... 4,4'-methylenebis(2-chloroaniline) ...

4,4'-Methylene-bis(2-chloroaniline) (MOCA) can produce tumors in rodents and dogs and an increased incidence of bladder tumors has been reported in exposed workers. It is therefore of interest to identify the human cytochrome P450 (P450) enzymes involved in MOCA N-oxidation, the primary reaction involved in the formation of an electrophilic product. Human liver microsomes were fractionated and MOCA N-oxidation activity was monitored through the procedure. The most active enzyme fraction corresponded to P450 3A4, as determined by immunochemical assays and N-terminal amino acid sequence analysis. Yeast recombinant P450 3A4 also had MOCA N-oxidation activity. Purified human liver P450 2A6 showed catalytic activity; however, anti-P450 2A6 inhibited less than 20% of the microsomal activity while anti-P450 3A4 inhibited up to 75%. Levels of marker activities of both P450 3A4 (nifedipine oxidation) and P450 2A6 (coumarin 7-hydroxylation) were measured in a set of human liver microsomes and both were correlated with MOCA N-oxidation rates. Gestodene and troleandomycin inhibited up to half of the microsomal MOCA N-hydroxylation activity but 7,8-benzoflavone showed only slight inhibition. Anti-P450 3A4 inhibited (up to 80% of) the microsomal transformation of MOCA to a product genotoxic as judged by bacterial SOS response. The work indicates that P450 3A4 makes a major contribution to human liver microsomal MOCA N-oxidation, and P450 2A6 has a minor role. P450 1A2, which catalyzes the hydroxylation of many arylamines, does not contribute to a great extent.

Following ip or oral administration of 4,4'-methylenebis 2-chloroaniline (MBOCA) to rats, approx 1/3 of the radioactivity was excreted in the urine. Only 1-2% was identified as MBOCA. At least 9 other metabolites were separated by reversed phase high performance liquid chromotography. Treatment of the urine with sulfatase-glucuronidase liberated 2 major deconjugated metabolites. Despite the extensive metabolism of MBOCA in the rat, analysis of urine of exposed workers showed only MBOCA at less than or equal to 1500 nmol/L. No evidence was found for the major metabolites identified in the rat.

Absorption, Distribution and Excretion · 5건

Studies that have examined the absorption, distribution, and excretion of MBOCA in experimental animals show that MBOCA is excreted rapidly and that only a small percentage of the administered dose is excreted unchanged.

There is no evidence of any major long-term retention of MBOCA in the rat, although 48 hours after oral administration of 10 mg radiolabeled MBOCA/kg body weight, approximately 2% of the administered radioactivity was associated with the liver.

In /an/ individual who was sprayed accidentally with molten MOCA, pharmacokinetic analyses (one-compartment model) indicated a biological half-time of 23 hr, with 94% elimination from the body in four days; 35% of the parent MOCA present in the urine was excreted as conjugates. High levels of MOCA (3.6 mg/L [13 m]) were also found in the urine of another worker accidentally sprayed with MOCA 4 and 11 hr after exposure. By 17 and 20 hr, the levels had decreased to 0.03-0.06 mg/L (0.1-0.2 M).

Percutaneous absorption of 4,4'-methylenebis(2-chloroaniline) was measured during dry contact exposure with organ cultures of neonatal foreskin. It was rapidly and progressively absorbed and passed through the skin without being metabolized.
