Reaction Steps
112 mg of the compound [22-1] was dissolved in 5 mL of tetrahydrofuran, and cooled to 0° C. Thereafter, 320 μL of methylmagnesium chloride (3.0M tetrahydrofuran solution) was added thereto, and the mixture was stirred for 1.5 hours while heating back to room temperature. To the reaction solution was added a saturated aqueous solution of ammonium chloride, and the solution was extracted with ethyl acetate. The obtained organic layer was washed with saturated brine and dried over anhydrous magnesium sulfate. The insolubles were filtered, the filtrate was concentrated under reduced pressure, and the resulting residue was purified by silica gel column chromatography (eluent: hexane/ethyl acetate=100/0-30/70), to obtain 53 mg of benzyl-4-[1-(4-{(1S)-1-[(tert-butoxycarbonyl)amino]ethyl}phenyl)-1-hydroxyethyl]piperidine-1-carboxylate [22-2] (hereinafter, referred to as the compound [22-2]).
