Reaction Steps

In a 50 ml recovery flask, 326.4 mg (0.639 mmol, 1.0 equivalent) of {(4-dipropylaminobutyl)-[4-(3-nitrobenzylidene)aminomethylbenzyl]amino}ethyl acetate (34) was dissolved in 3.3 ml of ethanol. The solution was cooled to 0° C. After the dropwise addition of 3.3 ml of a 2N hydrochloric acid aqueous solution over about five minutes, the mixture was reacted at 0° C. for three hours. After the addition of 3.3 ml of a 2N hydrochloric acid aqueous solution, the mixture was allowed to stand at 5° C. for 12 hours. The mixture was extracted five times with 25 ml of chloroform. The aqueous layer was added with 50 ml of chloroform and 40 ml of a 3N sodium hydroxide aqueous solution to make the mixture basic. The mixture was then subjected to phase separation and extracted three times with 25 ml of chloroform. The extract was dried over anhydrous sodium sulfate, filtered, and concentrated, thereby obtaining 2.09 g of [(4-aminomethylbenzyl)-(4-dipropylaminobutyl)amino]ethyl acetate (14) as a slightly yellow oily substance (yield: 79%).
