A mixture of 3-(benzyloxy)-N-(4-fluorobenzyl)-9,9-dimethyl-4-oxo-6,7,8,9-tetrahydro-4H-pyrazino[1,2-a]pyrimidine-2-carboxamide (prepared as described in Example 3, Step 1) (1 eq.), K2CO3 (2 eq.) and tert-butyl bromoacetate (3 eq.) in DMF was heated for 24 hours at 45° C. and then cooled to room temperature. Solvent was removed under reduced pressure and the residue was taken up in EtOAc and washed with brine, dried (Na2SO4), filtered and concentrated under reduced pressure to yield the ester. MS (ES) C30H35FN4O5 requires 550. Found: 551 (M+H+).
