Reaction Steps

To a solution of 1α,3β-bis(tert-butyldimethylsilyloxy) -20(S)-hydroxypregna-5,7,16-triene (400 mg, 0.72 mmol) in tetrahydrofuran (7.2 ml), sodium hydride (60% in oil, 46 mg, 1.15 mmol) was added and stirred at room temperature for 30 minutes under a nitrogen stream. After addition of N,N-dimethylacrylamide (308 mg, 3.11 mmol), the reaction mixture was further stirred at the same temperature for 3 hours, poured into saturated aqueous ammonium chloride, extracted with ethyl acetate (3 times) and washed with saturated aqueous sodium chloride and water. The organic layer was dried over anhydrous magnesium sulfate. After evaporation under reduced pressure to remove the solvent, the resulting residue was purified by column chromatography (hexane:ethyl acetate=2:1) to give the titled compound (459 mg, 96.9%) as a colorless oil.
