Reaction Steps
A mixture of 3-amino-3-(4-{3-methoxy-4-[3-(2-methylphenyl)ureido]phenylacetylamino}phenyl)-propanoic acid tert-butyl ester [53 mg, Reference Example 7(b)], butyraldehyde (14 mg), sodium acetate (16 mg) and 4 Å molecular sieves (0.2 g) in methanol (5 ml) was stirred at ambient temperature under an atmosphere of nitrogen and then treated with sodium cyanoborohydride (13 mg). After stirring for 1 hour the reaction mixture was treated dropwise with hydrochloric acid (0.5 ml, 1M) then poured into 10% aqueous potassium carbonate solution (10 ml). The mixture was extracted with ethyl acetate (30 ml). The organic phase was dried over magnesium sulphate and then evaporated. The residue was subjected to flash chromatography on silica gel eluting with ethyl acetate to give the title compound (44 mg) as a yellow foam.
