Reaction Steps

The compound obtained in Example 317 (3.66 g) is dissolved in 1,4-dioxane (45 ml), and thereto are added conc. hydrochloric acid (50 ml) and methanol (5 ml). The mixture is heated with stirring at 90° C. for 1.5 hour. To the mixture is added gradually a 2M aqueous sodium hydroxide solution (200 ml) under ice-cooling, and the mixture is neutralized with a saturated aqueous sodium hydrogen carbonate solution. The mixture is extracted with ethyl acetate, and the extract is washed, dried, and concentrated under reduced pressure. The residue is crystallized from diethyl ether to give 2-(4-aminophenyl)-4-(4-bromo-3,5-dimethoxyphenyl)-7-hydroxy-6-methoxy-3-methoxycarbonyl-1(2H)-isoquinolinone (2.54 g) as listed in Table 27.
