Reagents, catalysts & solvents 1
Procedure
To a solution of N-(4-(2-(2-(1H-imidazol-4-yl)phenoxy)ethyl)-3-chlorophenyl)-2-amino-2-(tetrahydro-2H-pyran-4-yl)acetamide (40 mg, 0.088 mmol) in dichloromethane (3 mL) was added pyridine (29 μL, 0.352 mmol) and acetyl chloride (29 μL, 0.264 mmol). The reaction was stirred at room temperature for 16 h. The solvent was removed under reduced pressure. The crude product was dissolved in methanol (4 mL) and refluxed for 2 h. The solution was concentrated and the residue was purified by flash column chromatography (silica gel, 5%-10% MeOH/DCM as eluent) to afford the desired product as a white solid (27 mg, 0.054 mmol, 61%). 1H NMR: 1.42-1.58 (m, 3H), 1.68 (d, 1H, J=12.8 Hz), 2.03-2.07 (m, 1H), 2.08 (s, 3H), 3.24-3.31 (m, 4H), 3.94 (t, 2H, J=11.2 Hz), 4.39 (t, 2H, J=6 Hz), 4.61 (t, 1H, J=8.4 Hz), 6.81 (d, 1H, J=8.4 Hz), 6.99 (d, 2H, J=8 Hz), 7.18-7.24 (m, 3H), 7.37 (dd, 1H, J=8 Hz, 1.6 Hz), 7.60 (s, 1H), 7.77 (s, 1H),
