Reaction Steps
2.1 1.5 ml of ammonia (7 M in methanol) are added under argon to 30 mg of “A1”, and the mixture is stirred in the microwave at 100°/10 bar for 40 minutes. 0.5 ml of ammonia solution (32%) and 5.06 mg of magnesium chloride are added, and the mixture is stirred at 120°/17 bar for a further 40 minutes. The mixture is subjected to conventional work-up, and the residue is chromatographed over an RP18 silica-gel column, giving 2.7 mg of 5-aminocarbonyl-3-benzyl-6-hydroxy-1H-indazole (“A3”).
