Stirred a solution of 3-Acetyl-1-{2-[2-methyl-4-(5-pyrimidin-2-yl-pyridin-2-yl)-piperazin-1-yl]-2-oxo-ethyl}-pyrrolidine-3-carboxylic acid [3-(4-fluoro-phenyl)-1Trityl-indazol-5-yl]-amide (10N) (600 mg, 0.664 mmol) in trifluoroacetic acid (20 ml) at room temperature overnight. The solvent was evaporated, water (100 ml) and 1N NaOH (10 ml) were added. The mixture was extracted with MeCl2 (2×150 ml), dried over MgSO4, filtered and solvent evaporated yielding a residue which chromatographed on silica gel eluting with 10% v/v MeOH/MeCl2 containing 2% NH4OH yielding product as a white solid.
