Reaction Steps
A stirred solution of 4-[5-(5-aminopyridin-2-yloxy)-1-methyl-1H-indole-2-carbonyl]piperazine-1-carboxylic acid tert-butyl ester (4.69 g, 10.4 mmol) in THF (250 mL) was ice cooled, then 3,4-dichlorobenzenesulfonyl chloride (3.7 g, 15.1 mmol) and pyridine (1.51 mL, 18.7 mmol) were added to the solution. The resulting mixture was stirred for 17 hours while gradually warming up to room temperature. Water was added to the mixture, and the aqueous layer was extracted with AcOEt. The extract was washed with saturated aqueous NaHCO3 and brine. The organic layer was dried over anhydrous magnesium sulfate and evaporated. The residue was purified by silica gel column chromatography (CH2Cl2:MeOH=40:1) to afford 5.26 g of the title compound as a pale yellow powder.
