Reagents, catalysts & solvents 1
Procedure
The crude product of EXAMPLE 428F (0.10 g) was treated with 1:1 dichloromethane/trifluoroacetic acid (10 mL). The reaction was stirred for 2 hours. The solvents were removed, and the residue was purified by reverse phase Gilson Prep HPLC system with a Phenomenex prep column (Luna, 5 μA, C18(2), 250×21.20 mm, 5 Å) eluting with 20-80% acetonitrile in water with 0.1% trifluoroacetic acid. Fractions containing product were concentrated, and the concentrate was diluted with dichloromethane, neutralized with aqueous NaHCO3, dried over Na2SO4, filtered, and concentrated. 1H NMR (500 MHz, dimethylsulfoxide-d6) δ 11.47 (s, 1H), 8.58 (t, 1H), 8.40 (d, 1H), 7.49-7.53 (m, 2H), 7.36 (d, 2H), 7.06 (d, 2H), 7.01 (d, 1H), 6.88 (t, 1H), 6.88 (d, 2H), 6.49 (d, 1H), 5.69 (s, 1H), 3.68-3.79 (m, 4H), 3.52-3.56 (m, 2H), 3.17 (br s, 4H), 2.81 (br s, 2H), 2.17-2.25 (m, 6H), 1.97 (s, 2H), 1.77-1.87 (m, 4H), 1.40 (t, 2H), 0.94 (s, 6H).
