Reaction Steps
A 250 mL 3-necked round bottom flask was charged with methyl 8-(4-(tert-butoxycarbonyl)piperazin-1-yl)-[1,2,4]triazolo[4,3-a]pyrazine-5-carboxylate (3.0 g, 8.3 mmol) and dry CH2Cl2 (30 mL). To the above was added dropwise a solution of 1.5 M diisobutylaluminum hydride in toluene (11 mL, 16.5 mmol) at −78° C. The resulting mixture was stirred at −78° C. for further 1 h then quenched by slow addition of methanol (10 mL). The mixture was poured into saturated aqueous NH4Cl (200 mL) and extracted with ethyl ether (100 mL×2). The combined organic layers were washed with saturated aqueous NaHCO3 (100 mL) and brine (100 mL), dried over anhydrous Na2SO4 and concentrated in vacuo. The residue was further purified by flash column chromatography on silica gel with 3% MeOH in CH2Cl2, to afford 1.4 g (50%) of the product as a white solid. MS m/z: 335 (M+H+).
