Reaction Steps
A mixture of ibrutinib (15 mg, 1.0 eq), (2,5-dioxopyrrolidin-1-yl)oxycarbonyloxymethyl acetate (1.5 eq), and NaHCO3 (3.0 eq) in 1:1 water and acetonitrile (0.5 mL) was stirred at 20° C. overnight. The reaction was then purified by reverse-phase (C-18) liquid chromatography using water and acetonitrile as eluents to yield compound (1). MS (ESI): m/z 557.2 (M+H)+.
