Reaction Steps

A solution of 4.5 g (13.4 mmol) of 4-[4-(Tetrahydro-pyran-2-yloxy)-but-1-ynyl]-piperidine-1-carboxylic acid tert-butyl ester and 1 g (4mmol) of pyrimidium toluene-4-sulfonate in 45 ml MeOH was stirred at 55° C. for 1 h. The reaction was partitioned between aqueous solution of 10% KHSO4 /ether (3×). The organic phases were washed with aqueous saturated NaHCO3, 10% NaCl, dried over Na2SO4 and evaporated to give 3.26 g (97%) of 4-(4-Hydroxy-but-1-ynyl)-piperidine-1-carboxylic acid tert-butyl ester, MS: 254 (MH+).
