Reaction Steps
1.38 g of 1,3-dimethyl-6-{4-[3-(2-methoxycarbonyl-4-nitrophenyloxy)propyl]piperazin-1-yl}-2,4(1H,3H)-pyrimidinedione (Compound 151--free form) were dissolved in 200 ml of methanol, followed by the addition of 100 ml of a 1.5 N aqueous solution of sodium hydroxide. The resultant mixture was stirred at 60° C. for 30 minutes. The reaction mixture was allowed to cool down, neutralized with dilute hydrochloric acid, and then concentrated to a total volume of 50 ml under reduced pressure. The concentrate was ice-cooled and precipitated crystals were collected by filtration, thereby obtaining 1.16 g of 1,3-dimethyl-6-{4-[3-(2-carboxy-4-nitrophenyloxy)propyl]piperazin-1-yl}-2,4(1H,3H)-pyrimidinedione.
