Procedure
Polyacrylamide Bio-Carriers (Bio-Rad Laboratories, Richmond, Calif.) with a swollen surface area of 16.5 m2 or the equivalent of 239 roller bottles (each having 690 cm2 available surface) were sterilized by autoclaving in aqueous buffer solution at pH 5.0 consisting of 0.05 M NaCl and 0.05 M 2(N-Morpholino)ethane sulfonic acid (MES). During autoclaving, the depth of the settled beads was less than two inches (<5.18 cm metric) and the temperature was maintained at 124° C. for about 40 to 60 minutes. After cooling, the beads were asceptically washed twice with phosphate buffered saline (PBS) and once with the aforesaid serum-containing medium. The PBS was prepared by dissolving 80 grams NaCl, 2 grams KCl, 2 grams KH2PO4 and 21.6 grams Na2HPO4.7H2O in 10 liters distilled water, followed by autoclaving.
