Procedure

To a solution of 3.7 g of t-butyl 4-(6-acetoxy-methylpyridine-2-carbonyl)piperazine-1-carboxylate in 10 mL of methanol was added 840 mg of potassium hydroxide and the mixture was heated to reflux for 4 hours. After the solvent was removed, water was added to the solution and the mixture was extracted with chloroform. The organic layer was washed with brine, dried over anhydrous magnesium sulfate, then the solvent was removed under reduced pressure. The residue was purified by chromatography on silica gel (methylene chloride-ethanol=50:1) to give 1.26 g of t-butyl 4-(6-hydroxymethylpyridine-2-carbonyl)piperazine-1-carboxylate as a colorless oil.
