Procedure

A 50-mL round-bottomed flask equipped with a reflux condenser was charged with 122b (310 mg, 1.0 mmol), 3-(acetoxymethyl)-2-(6-tert-butyl-8-fluoro-1-oxophthalazin-2(1H)-yl)pyridin-4-ylboronic acid 116c (1.24 g, 3.0 mmol), K3PO4 (424 mg, 2.0 mmol), sodium acetate (164 mg, 2.0 mmol), 1,1′-bis(diphenylphosphino)ferrocenedichloropalladium(II) (73 mg, 0.10 mmol), acetonitrile (10 mL), and water (0.5 mL). The system was subject to three cycles of vacuum/nitrogen flush and heated at 100° C. under N2 protection for 2.5 h. The reaction mixture was cooled to room temperature and concentrated under reduced pressure. The residue was diluted with dichloromethane (50 mL) and water (50 mL). The aqueous layer was separated and extracted with dichloromethane (3×20 mL). The combined organic layer was dried over Na2SO4, filtered, and concentrated under reduced pressure. The dark residue was purified with silica-gel column chromatography eluting with 60:1 dichloromethane/methanol to afford 122c (150 mg, 25%) as a yellow oil. MS-ESI: [M+H]+ 600.0
