2-aminonicotinic acid (7 g) and formamide (22.8 g) were heated at 167 C (internal temperature) for 2.5 hours. After cooling, the solid was recrystallized from 100 mL of hot water to give Pyrido[2,3-d]pyrimidin-4-ol as pale yellow powder (5.2 g, 69.7%). 1H NMR (DMSO, 400 MHz) δ 12.60 (br, 1H), 8.90 (br, 1H), 8.50 (m, 1H), 8.37 (s, 1H), 7.50 (m, 1H). Rt 0.87 min. MS (ESI+) [M+H]+148.
