Reaction Steps
To a stirred suspension of 10-(4-cyclohexyl-benzoyl)-10,11-dihydro-5H-pyrrolo[2,1-c][1,4]benzodiazepine-3-carboxylic acid of Example 22 (3.14 g) in dichloromethane (25 mL) was added oxalyl chloride (3.0 g). The reaction mixture was stirred for five hours at room temperature and then evaporated to dryness. The crude acid chloride thus obtained was dissolved in dichloromethane (50 mL) and added to a mixture of N,N-diisopropylethyl amine (1.0 g) and N-methyl piperazine (1.52 g) in dichloromethane (50 mL). The mixture was stirred at room temperature overnight, then washed with water and saturated aqueous sodium bicarbonate. The solution was dried over anhydrous sodium sulfate and then filtered through a short column of Magnesol®. Removal of the solvent followed by the addition of diethyl ether provided the title compound (2.50 g) as a pale yellow solid, m.p. 134–136° C.
