Reaction Steps

To a solution of (3R,7R,8aS)-7-hydroxy-3-(1H-indol-3-ylmethyl)-hexahydropyrrolo[1,2-a]pyrazine-1,4-dione (2.5 g) in dimethylformamide (30 mL) was added imidazole (1.7 g) and tert-butyl-chloro-diphenylsilane (4.35 mL). The resulting mixture was stirred overnight at room temperature and then partitioned between water and ethyl acetate. The organic layer was dried, filtered and concentrated in vacuo. The residue was purified by flash chromatography (SiO2, EtOAc) to afford (3R,7R,8aS)-7-(tert-butyl-diphenylsilanyloxy)-3-(1H-indol-3-ylmethyl)-hexahydropyrrolo[1,2-a]pyrazine-1,4-dione (4.6 g, 99%) (intermediate 7).
