Reaction Steps

A suspension of 2-methoxynicotinic acid (0.23 g; 1.5 mmol) in methylene chloride (15 ml) was treated with oxalyl chloride (0.14 mL; 1.6 mmol) and 2 drops of N,N-dimethylformamide. The reaction mixture was allowed to stir at room temperature for 30 minutes at which time triethylamine (0.3 mL; 2.2 mmol) and compound 1 (0.315 g; 1.66 mmol). After an additional 15 minutes of stirring the reaction mixture was washed with water and saturated aqueous sodium chloride, dried (magnesium sulfate) and concentrated. The crude residue was purified by column chromatography on silicia gel using 7:3 hexane:ethyl acetate as the eluent to give 0.37 g of 2-Methoxy-N-(1-phenyl-cyclohexylmethyl)-nicotinamide as a white solid. Mass Spec [M+H]+=325.
