Reaction Steps
To a solution of methyl 3-[2-(N-methylamino)-4-phenyl-5-thiazolyl]propionate (400 mg) in N,N-dimethylformamide (10 ml) was added sodium hydride (60% in oil, 70 mg) at 0° C. and the mixture was stirred at said temperature for 10 min. 4-(4-Chloromethylphenoxymethyl)-5-methyl-2-phenyloxazole (500 mg) was added and the mixture was stirred at room temperature for 1 hr. The reaction mixture was poured into ice water, neutralized with 2N hydrochloric acid and extracted with ethyl acetate. The ethyl acetate layer was washed with water, dried (MgSO4) and concentrated. The residue was subjected to silica gel column chromatography, and methyl 3-[2-[N-methyl-N-[4-(5-methyl-2-phenyl-4-oxazolylmethoxy)benzyl]amino]-4-phenyl-5-thiazolyl]propionate (580 mg, yield 72%) was obtained as a colorless oil from fraction eluted with ethyl acetate-hexane (1:9, volume ratio).
