Procedure
A mixture of N-benzyl-2-(butyrylamino)-3-furamide (1-2, 1 equiv) and sodium hydroxide (0.1 equiv) in ethylene glycol is heated at 130° C. for 5 h. The reaction mixture is allowed to cool, then partitioned between a half-saturated aqueous sodium chloride solution and ethyl acetate. The combined organic layers are dried over sodium sulfate and concentrated. The residue is purified by flash column (hexanes initially, grading to 100% ethyl acetate) to provide 3-benzyl-2-propylfuro[2,3-d]pyrimidin-4(3H)-one (1-3).
