Reaction Steps

(2R)-3-Methyl-2-{[4-(trifluoromethyl)phenyl]amino}butan-1-ol (1.67 g, 6.76 mmol) obtained in Step 1 was dissolved in methylene chloride (30 mL) and triethylamine (1.20 mL, 8.67 mmol), and methanesulfonyl chloride (0.63 mL, 8.14 mmol) was added dropwise under cooling. After stiring overnight, the mixture was worked-up according to a conventional method to give N-[(1R)-1-(chloromethyl)-2-methylpropyl]-4-(trifluoromethyl)aniline as a crude product. The obtained crude product was dissolved in dimethylformamide (20 mL), sodium azide (0.40 g, 6.15 mmol) was added, and the mixture was stirred overnight at 80° C. The solvent was evaporated, and the residue was worked-up according to a conventional method to give a crude product. The obtained crude product was dissolved in ethyl acetate (20 ml), 10% palladium/carbon (catalytic amount) was added, and the system was purged with hydrogen. The mixture was stirred overnight at room temperature, palladium/carbon was filtered off, and the solvent was evaporated to give the title compound (1.50 g, 6.09 mmol, 90%).
