Reaction Steps

To a solution of 1-benzyl-4-(5-chloro-2-methoxy-phenyl)-piperidin-4-ol (4b, 17 g, 0.05 mol) in dry dichloromethane (70 mL) was added dropwise DAST (10 g, 0.061 mol) at −78° C. under N2 atmosphere. The mixture was stirred at this temperature for 1 h and then warmed to room temperature slowly. NaHCO3 (sat., aq., 200 mL) was carefully added dropwise to quench the reaction. The separated aqueous was extracted with dichloromethane (150 mL×3), the combined extracts was washed with water and brine, dried over Na2SO4, concentrated to dryness. The residue was titrated with petroleum ether and the precipitated solid was filtered and washed with petroleum ether to afford 1-benzyl-4-(5-chloro-2-methoxy-phenyl)-4-fluoro-piperidine 4c as white powder.
