Reaction Steps
4,6-Dichloro-5-fluoro-2-(methylthio)pyrimidine (see Intermediate E) (0.422 g, 1.991 mmol), commercially available (1,3-thiazol-2-ylmethyl)amine hydrochloride (0.300 g, 1.992 mmol) and triethylamine (0.61 mL, 4.3765 mmol) were dissolved in THF (3 mL) and left to stir for 4 days. The reaction mixture was diluted with water, and the aqueous layer was extracted with ethyl acetate. The organics were dried (MgSO4) and concentrated to produce 6-chloro-5-fluoro-2-(methylthio)-N-(1,3-thiazol-2-ylmethyl)-4-pyrimidinamine as a yellow-orange solid (0.6773 g). LCMS: (M+H)+=291.0.
