Reaction Steps
Sodium hydride (60% dispersion in oil, 5 mg) was added at room temperature under an atmosphere of nitrogen to a solution of N-[4-(1,3-benzodioxol-5-yl)-3-methoxy-1H-pyrazol-5-yl]-2-phenyl-1-ethanesulfonamide (Preparation 56) in anhydrous tetrahydrofuran (2.5 ml). After 10 minutes, bromomethyl methyl ether (12 μl) was added and the reaction was stirred overnight. An aqueous saturated solution of ammonium chloride (4 ml) was added and the mixture was extracted with ethyl acetate (3×6 ml). The combined organic fractions were washed with brine (6 ml), dried over sodium sulfate and concentrated under reduced pressure. The oily residue was purified by column chromatography (Sep-Pak)™ eluted with hexane:ethyl acetate (4:1) then with hexane:ethyl acetate (1:1) to yield the title compound as a white solid (40 mg).
