Reaction Steps
The product from Example 279A was treated with 2N NaOH aqueous solution (1 mL), ethyl alcohol (0.5 mL) and tetrahydrofuran (0.5 mL) at room temperature overnight. 1N hydrochloric acid was dropped to the reaction mixture until pH=2˜3. Ethyl acetate (3 mL) was used to extract the product. The organic layer was separated, dried (Na2SO4), concentrated in vaccuo, and purified by HPLC to provide the titled compound. 1H NMR (500 MHz, DMSO-d6) δ12.03-12.70 (br.s, 1 H), 8.95 (s, 1 H), 7.24 (m, 8 H), 7.04 (m, 1 H), 6.94 (m, 4 H), 6.69 (m, 1 H), 6.49 (m, 2 H), 4.06 (s, 2 H), 4.03 (s, 2 H), 3.94 (t, J=6.4 Hz, 2 H), 3.82 (m, 1 H), 3.72 (m, 1 H), 3.31 (dd, J=13.6, 9.2 Hz, 1 H), 2.98 (t, J=11.1 Hz, 1 H), 2.91 (s, 3 H), 2.38 (m, 5 H), 2.24 (m, 1 H), 1.90 (m, 3 H), 1.65 (m, 1 H), 1.52 (m, 1 H), 1.34 (m, 1 H); MS (APCI+) m/z 686.1 (M+H)+.
