Reaction Steps

To 671.4 mg of 6-methoxy-2-[4-[2-(1-pyrrolidinyl)-ethoxy]phenyl]benzo[b]thiophene (Example 1, Part B) in 10 mL of 1,2-dichloroethane was added 1.114 g of AlCl3 at 0° C., followed by dropwise addition of 0.30 mL of 4-fluorobenzoyl chloride. The deep red solution was stirred at 0° C. for 1 h and then at 0 to 15° C. for 19 h. The reaction was quenched by pouring the reaction mixture into 50 mL of ice-cold 2.0 N NaOH solution. The mixture was then extracted with 3×100 mL of EtOAc which was washed with 50 mL of H2O and brine. Combined organic layers were dried over MgSO4, concentrated, and purified by flash chromatography with 5 v/v % (10% conc NH4OH in MeOH) in CH2Cl2 to give 826.4 mg (92%) of the product ketone as viscous oil.
