Reaction Steps
Phosphorus oxychloride (599 μL, 6.42 mmol) was added dropwise to dry N,N-dimethylformamide, (497 μL, 6.42 mmol) in toluene (0.6 mL) at 0° C. The mixture was stirred for 5 minutes. Then 4-(1-(4-methoxyphenyl)-3-propyl-1H-pyrazol-5-yl)-3,5-dimethylisoxazole (160 mg, 0.51 mmol) was added, dissolved in toluene (0.5 mL) and the reaction mixture was heated at 150° C. for 20 minutes in the microwave. CH2Cl2 and water were added, a phase separator was used to separate and dry the organic phase. Filtration through a short silica plug gave semi-crude 5-(3,5-dimethylisoxazol-4-yl)-1-(4-methoxyphenyl)-3-propyl-1H-pyrazole-4-carbaldehyde which was used as such in the next step without further purification.
