5-chloro-1-isobutyl-6-(4-methoxybenzyl)-3-methyl-1H-pyrrolo[3,4-d]pyrimidine-2,4(3H,6H)-dione (14 mg, 0.025 mmol) is placed in a Biotage microwave tube, and then aniline (0.5 mL, 5.4 mmol) is added. The mixture was heated at 180° C. for 4 h. The mixture is purified by a semi-preparative HPLC to give 13 mg of pure product (yield: 81%). MS (ESI) m/z 433.2 [M+H]+.
