Reaction Steps

A stirred mixture of N-[(6-bromopyridin-2-yl)-(4-fluorophenyl)-methyl]-formamide (6.4 g, 21 mmol) in POCl3 (41 mL) is warmed at reflux. After 1 hour, the reaction mixture is cautiously quenched by a slow addition to saturated aqueous NaHCO3 and the mixture is extracted into EtOAc (3×200 mL). The combined organic layers are dried over Na2SO4 and concentrated. The crude material is purified by silica gel chromatography eluting with 30% EtOAc in hexanes to afford 5-bromo-1-(4-fluorophenyl)-imidazo[1,5-a]pyridine.
