Reaction Steps
To a suspension of sodium hydride (60% in oil, 37 mg) in tetrahydrofuran (2 mL) were added a solution of tert-butyl {[5-(2-fluoropyridin-3-yl)-4-fluoro-1H-pyrrol-3-yl]methyl}methylcarbamate in tetrahydrofuran (1 mL), 15-crown-5 (205 mg) and 3-thiophenesulfonyl chloride (136 mg) at room temperature and the mixture was stirred for 2 hr. The reaction mixture was diluted with water, and extracted with ethyl acetate. The separated aqueous layer was extracted again with ethyl acetate. The combined organic layer was washed with saturated brine, dried over anhydrous magnesium sulfate, and concentrated under reduced pressure. The residue was purified by basic silica gel column chromatography (eluent: hexane-ethyl acetate=6:1→3:2) to give the title compound as a pale-yellow oil (yield 264 mg, 91%).
